Positron-emitting radioligands for imaging neuroleptic receptors

Description: A series of /sup 18/F-labeled butyrophenones (benperidol, haloperidol, spiroperidol and N-methylspiroperidol) were evaluated in baboons and rats with respect to potential utility as radioligands for studying neuroleptic receptors in the living human brain by positron emission tomography. These compounds were administered to baboons, and the radioactivity distributions to the striatum, and to the cerebellum were determined by PET at times up to 8 hours after isotope injection. 4 refs. Radiopharmaceuticals for diagnosis and treatment

Description: We report on our efforts in PET basic radiochemistry, radiopharmaceutical synthesis, and preclinical radiopharmaceutical evaluation. These efforts are focused on three fronts. First predictive abilities in nucleophilic aromatic substitution with [[sup 18]F]fluorination of substituted aromatic rings. Although radiochemical yields can be predicted within a very similar group of compounds with similar leaving groups and substituent patterns, generalization to all nucleophilic substitutions with [[… more

Radiotracers in PETT: strategies for in vivo receptor activity, Schizophernia, and Alzheimer's Dementia studies

Description: Using /sup 18/F-spiperone, a one compartment system with a driving function as model, blocking agents such as butaclamol and ketanserin, assay of the live adult female baboon striatum over the 8 h period, and assay of the parent compound in plasma, it is apparent that residence times in the living tissue and those estimated from in vitro tritium data are at variance. Occupancy rises to a maximum for /sup 18/F benperidol and /sup 18/F haloperidol after approx. 25 minutes and for /sup 18/F spiper… more

Chiral separation of pharmaceutical compounds using electrochemically modulated liquid chromatography (EMLC)

Description: This research explores the application of a new technique, termed electrochemically modulated liquid chromatography (EMLC), to the chiral separations of pharmaceutical compounds. The introduction section provides a literature review of the technique and its applications, as well as brief overview of the research described in each of the next two chapters. Chapter 2 investigates the EMLC-based enantiomeric separation of a group of chiral benzodiazepines with {beta}-cyclodextrin as a chiral mobil… more
