Determination of somatic mutations in human erythrocytes by cytometry

Description: Flow cytometric assays of human erythrocytes labeled with monoclonal antibodies specific for glycophorin A were used to enumerate variant cells that appear in peripheral blood as a result of somatic gene-loss mutations in erythrocyte precursor cells. The assay was performed on erythrocytes from 10 oncology patients who had received at least one treatment from radiation or mutagenic chemotherapy at least 3 weeks before being assayed. The patients were suffering from many different malignancies (… more

Cytometric analysis of mammalian sperm for induced morphologic and DNA content errors

Description: Some flow-cytometric and image analysis procedures under development for quantitative analysis of sperm morphology are reviewed. The results of flow-cytometric DNA-content measurements on sperm from radiation exposed mice are also summarized, the results related to the available cytological information, and their potential dosimetric sensitivity discussed. Single particle analysis with a 360/sup 0/ light scattering photometer

Description: Light scattering by single spherical homogeneous particles in the diameter range 1 to 20 ..mu..m and relative refractive index 1.20 is measured. Particle size of narrowly dispersed populations is determined and a multi-modal dispersion of five components is completely analyzed. A 360/sup 0/ light scattering photometer for analysis of single particles has been designed and developed. A fluid stream containing single particles intersects a focused laser beam at the primary focal point of an ellip… more

Flow cytogenetics: Fundamentals and new developments

Description: The purpose of this report is to provide (1) an introduction to flow cytogenetics and (2) resource information for additional and more detailed information for those interested in either using or further developing the technology. Several recent reviews are available and provide in-depth information on all aspects of flow cytogenetics, this information will by and large, not be repeated here. Examples of common types of flow cytogenetic data are included along with an explanation of their inter… more

Optimization of the Mini-Flo flow cytometer

Description: A new method of collecting light scattering from a liquid flow cytometer has been proposed; this apparatus is named the Mini-Flo flow cytometer. The Mini-Flo uses a high numerical aperture collection immersed in the flow stream. The collector consists of a conically tipped fiber optic pipe and terminating optical detector. This study was performed to improve the signal/noise ration and optimize the Mini-Flo`s performance for HIV blood detection applications. Experiments were performed to gauge … more
