Radiation toxicology

Description: The extensive studies on both human and experimental animal populations have provided information that allows radiation protection standards to be set with greater confidence than for most if not all other carcinogenic agents. Furthermore, both international and national advisory bodies are continually updating the risk estimates and the standards as new information is available. However, it is clear that we need models that take into account the multistage nature of carcinogenesis. Studies in … more

Effects of chemical carcinogens of hemopoiesis, immunopoiesis and viral oncogenesis. Technical progress report, December 1, 1977--September 30, 1978. [Mechanisms of potentiation of viral leukemogenesis by MMS, benzopyrene, and DMBA]

Description: During the past year we have concentrated on defining the circumstances under which methyl methanesulfonate (MMS), benzo(a) pyrene (BP), and 7,12-dimethylbenz(a)anthracene (DMBA) interact with Friend virus (FLV) to produce leukemia. The optimum scheduling for each and also the effective dose levels of the chemicals have been partially determined. There are at least three critical factors which govern whether or not a leukemogenic interaction can be shown between the chemical agents and the viru… more

Effect of chemical carcinogens on hematopoietic stem cells: temporal correlation between methyl methane sulfonate induced depression of CFU-S and potentiation of Friend viral leukemogenesis in vivo. [Mice]

Description: Methyl methane sulfonate (MMS) was shown to reduce the number of transplantable pluripotent hematopoietic stem cells (CFU-S) in the mouse spleen. A mild depression in CFU-S numbers was seen as early as 1 hr after injection of the chemical carcinogen. Maximal depression was found to be at 5 hrs, followed by a recovery of up to 75% of normal levels by 24 hrs. Comparison of the data with earlier reslts which demonstrated a potentiating effect of MMS on viral leukemogenesis in vivo revealed a good … more

Test for detection of clustering over time. [Statistical study of spontaneous abortions in NYC]

Description: The use of the scan statistic in testing for clustering in time is discussed with particular reference to a reported clustering of trisomies in spontaneous abortions in New York City (spontaneous abortions have been used on occasion to monitor environmental health effects). The scan statistic is the maximum number of observed cases in an interval of preselected length, as the interval is allowed to scan, or slide along, the time frame of interest. A table of the lower tail of the distribution o… more

Effects of rainfall acidification on plant pathogens

Description: Wind-blown rain, rain splash, and films of free moisture play important roles in the epidemiology of many plant diseases. The chemical nature of the aqueous microenvironment at the infection court is a potentially significant factor in the successful dissemination, establishment, and survival of plant pathogenic microorganisms. Acidic rainfall has a potential for influencing not only the pathogen, but also the host organism, and the host-pathogen complex. Although host-pathogen interactions add… more

Behavior as a sentry of metal toxicity

Description: Many of the toxic properties of metals are expressed as behavioral aberrations. Some of these arise from direct actions on the central nervous system. Others arise from primary events elsewhere, but still influence behavior. Toxicity may be expressed either as objectively measurable phenomena, such as ataxia, or as subjective complaints, such as depression. In neither instance is clinical medicine equipped to provide assessments of subtle, early indices of toxicity. Reviewers of visual disturba… more

Brain plasticity, memory, and aging: a discussion

Description: It is generally assumed that memory faculties decline with age. A discussion of the relationship of memory and aging and the possibility of retarding the potential decline is hampered by the fact that no satisfactory explanation of memory is available in either molecular or anatomical terms. However, this lack of description of memory does not mean that there is a lack of suggested mechanisms for long-term memory storage. Present theories of memory usually include first, neurophysiological or e… more

Lymphocyte blastogenesis in the diagnosis of tuberculosis in cattle

Description: Considerable progress has been made towards the eradication of tuberculosis in cattle in the United States. No reliable blood test has been developed for detecting infected animals. An in vitro test for tuberculosis must have a high level of sensitivity with adequate specificity to differentiate infections due to Mycobacterium bovis and those caused by other mycobacteria. Moreover, it would be desirable that the test system be simple ad readily automated. The accuracy of lymphocyte immunostimul… more

Genetic influence on hypertension induced by cadmium in Dahl hypertension-resistant and hypertension-sensitive rats

Description: Dahl hypertension-resistant (R) and hypertension-sensitive (S) rats, with opposite genetically controlled propensities for hypertension, were used to determine if cadmium-induced hypertension was also dependent upon genetic predisposition. When intraperitoneal injections of cadmium (1 and 2 mg/kg) were given to male and female R and S rats maintained under ordinary laboratory conditions, female S rats manifested a significant increase in blood pressure which persisted for 16 weeks, whereas the … more

Inborn anemias in mice. Progress report to accompany twenty-first renewal proposal, 1 May 1975--30 April 1976

Description: Progress is reported on studies on hereditary anemias of mice. At present under study are four macrocytic anemias, four hemolytic anemias, nonhemolytic microcytic anemia, transitory siderocytic anemia, sex-linked iron-transport anemia, and the autoimmune hemolytic anemia of NZB. Each of these blood dyscrasias is caused by the action of a unique mutant gene, each of which determines the structure of different intracellular molecules, and thus controls a different metabolic process. Thus our wide… more

Environmental contaminants as origins of disordered behavior

Description: Behavioral toxicology studies the behavioral effects of contaminants, such as heavy metals, in environmental and occupational settings. A classic example of metal poisoning with behavioral effects is Pink Disease, or acrodynia, due to mercurous chloride intoxication in children. Thalidomide is a more prominent example. Behavioral changes, unlike tangible consequences of pollution, are difficult to perceive as dangers and to correct. Other examples of environmental pollutants causing behavioral … more

Tumorigenic action of beta, proton, alpha, and electron radiation on the rat skin. Progress report, August 1, 1977--January 31, 1979

Description: Epithelial tumors are induced reproducibly in rat skin exposed to ionizing radiation, ultraviolet light, or polycyclic aromatic hydrocarbons. For these carcinogens, the comparative risk of tumor formation is being examined for clues to the generality of action and the mode of interaction of diverse carcinogens. Molecular lesions, such as, pyrimidine dimers and single-strand breaks in DNA have been measured as a basis for assessment of cellular dose. Dose-response for single doses, time response… more

Tissue Enzyme Changes in Parabiotic Rats with Subcutaneous Lymphoma

Description: A solid lymphoma was implanted into normal rats and into one partner of parabiotic pairs. Appreciable decreases in hepatic ornithine aminotransferase and arginase occurred about a week earlier (4 to 6 days after implantation) in single than in parabiotic hosts. By 18 to 21 days significant decreases in both enzymes were also apparent in the host partner. The hepatic thymidine kinase showed a 5-fold elevation in single hosts 4 days after implantation; by 14 days its levels were about 200 times a… more

Impact of thermal loading and other water quality parameters on the epizootiology of Epistylis and Aeromonas infections of centrarchids. Progress report, September 1, 1976--July 30, 1977

Description: During the past year, mounting evidence has been accumulated which indicates that the causative agent for red-sore disease among bass is the bacterium, Aeromonas hydrophila, and not the ciliate, Epistylis, as suggested in previous literature. Red-sore disease among bass in Par Pond continues to be highly seasonal with peak periods of infection occurring in spring and summer, with declining levels in fall and summer. The disease continues to be more prevalent among bass from thermally altered ar… more
