Abstract: This presentation discusses research on the hypoxic effects in contraction of the ductus arteriosus (DA) in the chicken embryo. The authors' goal is to understand the role of Rho-kinase in the oxygen-induced contraction that occurs at hatching: Ductus smooth muscle contracts when the myosin light-chain to produce relaxation; and Rho-kinase mediates an oxygen-induced contraction by inhibiting the action of myosin phosphate-mediated dephosphorylation of myosin light-chain, thus promoting contraction. The authors examined the role of Rho-kinase in ductus contraction by exposing ductus vessels to a Rho-kinase inhibitor (Y-27632) after being pre-contracted by oxygen.
