Immunofluorescence as a Method for the Rapid Identification of Streptococcus Faecalis in Water

Description: The development and refinement of FA has been adequately investigated with major emphasis on pathogenic microorganisms. The development of this technique has reduced both the time and number of biochemical tests necessary to identify a diversity of organisms. The organisms included are the protozoans, as described by Goldman (1953 and 1957) and by Ingram (1961), viruses, as reported by Liu (1955a) and Burgdorfer and Lackman (1960a), pathogenic bacteria which have been investigated by Moody, Goldman, and Thomason (1956), Moody and Winter (1959), Deason, Falcone, and Harris (1957) and Thomason, Cherry, and Moody (1957). Various fungi have been studied with FA by Kaufman and Brandt (1964), Kaufman and Kaplan (1961 and 1963) and Gordon (1958). Therefore, due to the success of the fluorescent antibody technique in many areas of microbiology in previous investigations, the logical assumption was that immunofluorescence might be incorporated into an efficient system in which a specific organism associated with fecal pollution, such as S. faecalis, could be rapidly identified. Based on this assumption, the feasibility of fluorescent antibody techniques, using S. faecalis was investigated as a means of rapid determination of bacterial pollution in water. Although much progress has been achieved in the study of cytochemical reactions by immunofluorescence, no attention has been focused on the application of this method as a determinative tool by which water contamination, due to the presence of the enterococci, could be demonstrated. Specifically, the purpose of the research reported in this dissertation was to devise an applicable, valid, and rapid method that could be employed in the detection and identification of S. faecalis.

A Musical and Dramatic Analysis of the Principal Tenor Roles in Mozart's Singspiele

Description: This paper will examine one area of Mozart's work, the Singspiele. This study is an analysis of the principal tenor roles of Mozart's Singspiele. The organization for analyzing these works conforms to three periods in Mozart's life. (1) Childhood and Early Youth, to 1774; (2) The Period of the First Masterworks, 1774-1781; and (3) The Years in Vienna, 1781-1791. Related biographical data and historical background have been utilized in discussing each work. Because the Singspiele is a musical composition, analyses will consider music as the major source of development, using plot and character wholly as supporting features.

Contemporary Music Studies for the Concert Band

Description: The purpose of this thesis is to isolate the techniques presently being used by composers and to define and explain these techniques. The thesis concludes with a series of studies based on these twentieth-century compositional techniques for high school students in the form of warm-up and technique materials. The purpose of this study was to devise a sequence of studies designed to acquaint band members with twentieth-century composition techniques found in contemporary band literature.

A Method of Filming a Music and Fine Arts Program

Description: In reviewing the events of the advances of science and technology of the past two decades one is struck by the magnificence of man's accomplishments. The first moon walk televised live in 1969, the widespread utilization of color television receivers, the educational uses of sophisticated audio-visual equipment such as overhead projectors, slide projection, sound tape recorders, close-circuit television and videotape and the almost omniscient usage of television, is ample evidence to demonstrate that the tools necessary to accomplish a task are readily accessible items in this age. We are introduced to new gadgets, innovations, and events (like the moon walk) at such a rapid pace that we have become more flexible and more receptive to change; so much so that at times many of us are even blase about nearly miraculous accomplishments.

The Relationship of Children's Perception of Parental Punitiveness toward Aggression and their Church Attendance

Description: One of the main purposes of the present study was to use a parental punitiveness scale, that was developed on the assumption that parental punitiveness is a function of the situation in which aggression takes place. This in turn was used to determine what relationship a child's perception of parental discipline toward aggression has to varying degrees of church attendance of the child.

Food, Feeding Selectivity, and Ecological Efficiencies of Fundulus notatus (Rafinesque) (Osteichthyes; Cyprinodontidae)

Description: This study was made to further define the trophic dynamics of Fundulus notatus by determining its ration composition under natural conditions, measuring feeding selectivity under various laboratory conditions of prey-species composition and availability, and determining the efficiencies with which F. notatus utilizes ingested chironomid larvae.

The Effect of Teaching Beginning College Mathematics by Television

Description: The purposes of this study were (1) to compare the achievement levels of students enrolled in a beginning college mathematics course when taught by (a) closed-circuit television followed by student-assisted study periods, (b) closed circuit television followed by access to videotape replay with no supervised study periods, (c) closed-circuit television followed by unsupervised study and discussion, and (d) regular lecture-recitation methods conducted by the television instructor, and (2) to ascertain the students' attitudes toward their instructor, course, and method of instruction.

Alto Requiem

Description: Alto Requiem is a requiem mass for contralto, chorus, and organ. Several unifying elements have been used throughout the composition. Most of the homophonic texture is limited to quartal and secundal structures. The melodic material is also predominantly quartal and secundal. Key relationships between and within movements are limited to related major and minor modes, modes a major or minor second apart, and modes a fourth apart. Further unity is achieved by emplying motives from the chants of the Roman Missa pro defunctis. These chants are used in the vocal lines as well as in the organ accompaniment.
