Studies of human mutation rates

Description: The three objectives of the program are: To isolate by the technique of two-dimensional polyacrylamide gel electrophoresis (2-D PAGE), proteins of special interest because of the relative mutability of the corresponding gene, establish the identity of the protein, and, for selected proteins, move to a characterization of the corresponding gene; To develop a more efficient approach, based on 2-D PAGE, for the detection of variants in DNA, with special reference to the identification of a variant… more

Molecular mapping of chromosomes 17 and X

Description: Progress toward the construction of high density genetic maps of chromosomes 17 and X has been made by isolating and characterizing a relatively large set of polymorphic probes for each chromosome and using these probes to construct genetic maps. We have mapped the same polymorphic probes against a series of chromosome breakpoints on X and 17. The probes could be assigned to over 30 physical intervals on the X chromosome and 7 intervals on 17. In many cases, this process resulted in improved ch… more

In situ measurement of electromigration-induced transient stress in Pb-free Sn-Cu solder joints by synchrotron radiation based X-ray polychromatic microdiffraction

Description: Electromigration-induced hydrostatic elastic stress in Pb-free SnCu solder joints was studied by in situ synchrotron X-ray white beam microdiffraction. The elastic stresses in two different grains with similar crystallographic orientation, one located at the anode end and the other at the cathode end, were analyzed based on the elastic anisotropy of the {beta}-Sn crystal structure. The stress in the grain at the cathode end remained constant except for temperature fluctuations, while the compre… more

Magnetic bead purification of labeled DNA fragments forhigh-throughput capillary electrophoresis sequencing

Description: We have developed an automated purification method for terminator sequencing products based on a magnetic bead technology. This 384-well protocol generates labeled DNA fragments that are essentially free of contaminates for less than $0.005 per reaction. In comparison to laborious ethanol precipitation protocols, this method increases the phred20 read length by forty bases with various DNA templates such as PCR fragments, Plasmids, Cosmids and RCA products. Our method eliminates centrifugation … more

METAL ION ANALYSIS USING NEAR-INFRARED DYES AND THE ''LABORATORY-ON-A-CHIP''

Description: The DOE currently has the daunting task of deactivating 7,000 contaminated buildings and decommissioning 900 contaminated buildings that remain from the United States' involvement in nuclear weapons development over the last 50 years.1 In addition to decontaminating the metal and concrete building materials that comprise these building structures, this program will require the decontamination and disposal of more than 180,000 metric tons of scrap metal.1 The DOE is also currently entrenched in … more

Attomole quantitation of protein separations with accelerator mass spectrometry

Description: Quantification of specific proteins depends on separation by chromatography or electrophoresis followed by chemical detection schemes such as staining and fluorophore adhesion. Chemical exchange of short-lived isotopes, particularly sulfur, is also prevalent despite the inconveniences of counting radioactivity. Physical methods based on isotopic and elemental analyses offer highly sensitive protein quantitation that has linear response over wide dynamic ranges and is independent of protein conf… more

Protection of Graphite by Impregnation. Quarterly Progress Report No. 3 for September 1-November 30, 1960. Vitro Job 2195

Description: The hypothesis that sample resistivity is the controlling factor in electrokinetic impregnation was substantiated. Experiments show that dense clay- bonded silicon carbide can be impregnated beneath the surface pores whereas only surface pore impregnation is achieved with both dense and porous graphite. The resistivity of the materials differ by a factor of about 1O/sup 10/. Although deep penetration of the substrate by the particles of the coating system is not achieved during deposition, ther… more
