Enhancement of photoassimilate utilization by manipulation of the ADPglucose pyrophosphorylase genes. Progress report, [April 15, 1990--April 14, 1991]

Description: The long term goal of this project is to assess the feasibility of increasing the conversion of photosynthate a key regulatory enzyme in starch biosynthesis. In developing storage tissues such as cereal seeds and tubers, starch biosynthesis is primarily regulated by the gene activation, expression, and allosteric regulation of ADPglucose pyrophosphorylase, as well as starch synthase, and branching enzyme. During the last year we have elucidated the structure of both subunits which compose this … more

Areally averaged estimates of surface heat flux from ARM field studies

Description: The determination of areally averaged surface fluxes is a problem of fundamental interest to the Atmospheric Radiation Measurement (ARM) program. The Cloud And Radiation Testbed (CART) sites central to the ARM program will provide high-quality data for input to and verification of General Circulation Models (GCMs). The extension of several point measurements of surface fluxes within the heterogeneous CART sites to an accurate representation of the areally averaged surface fluxes is not straight… more

Enhancement of photoassimilate utilization by manipulation of the ADPglucose pyrophosphorylase gene. Progress report, [April 15, 1987--April 14, 1988]

Description: Many agronomically important crops are viewed as significant resources of renewable energy. Overall crop productivity could be increased if the efficiency of photoassimilate conversion into dry matter such as starch were improved in storage tissues. Starch production is controlled by the catalytic activity of ADPglucose pyrophosphorylase in the first step of starch biosynthesis. This research focuses on the genetic structure and molecular mechanisms by which it is controlled during plant develo… more

Feasibility studies for the Sonic Sensor System for noninvasive temperature measurement in the food processing industry

Description: Feasibility studies for development of the Sonic Sensor System to measure temperature in the interior of particulates during food processing are described. The method involves tomography-based acquisition and interpretation of speed of sound data. The method is found to be feasible, and representative engineering problems and other technical issues are identified. A two-step approach to concept commercialization, involving replacement of the present puncture-and-thermometer method is convention… more

How to Increase the Potato Crop by Spraying

Description: "This bulletin tells how to control the Colorado potato beetle and late-blight (the worst two enemies of the potato), blister beetles, flea-beetles, cutworms, and other caterpillars, leaf-hoppers, "aphis" or plant-lice, early-blight, and other foliage diseases. For most of these insects and for the diseases discussed in this bulletin, spraying with Bordeaux mixture and arsenate of lead is recommended, but for leafhoppers and plant-lice, contact sprays, such as nicotine sulphate and eumulsions, … more

The Sweet-Potato Weevil and Its Control

Description: "Immense losses of sweet potatoes in the Gulf States are being caused by the sweet-potato weevil. This foreign pest, introduced into the United States years ago, has become very destructive recently and now threatens to invade all States in which sweet potatoes are grown. The slender, metallic-blue weevil, about a quarter of an inch long with red legs and 'waist,' attacks leaves, stems, and roots or 'tubers,' and its whitish larvae or grubs tunnel the stalks and roots and inflict great damage, … more

[Induction of polygalacturonases important in pathogenicity of Pseudomonas solanacearum]

Description: Recent studies on the importance of hydroxyproline-rich glycoproteins (HPRG`s) in the nature and function of plant cell walls have led to the question as to whether proteolytic enzymes are also involved in tissue maceration and act in concert with other cell wall degrading enzymes in the process. The primary objective of this research was to determine whether proteolytic enzymes, in combination with other enzymes, are involved in the degradation of plant cell walls and thus may be essential for… more

[Enhancement of photoassimilate utilization by manipulation of the ADPglucose pyrophosphorylase gene]. Progress report, April 1993--January 1994

Description: Part I of this research concerns patterns of gene expression of ADPG-PP in native and transgenic potato plants. The expression of both potato ADPG-PP subunits were analyzed on the transcript and antigen levels. The small and large subunits were coordinately expressed during tuber development suggesting a role for the temporal regulation of ADPG-PP expression as well as providing further support for earlier work in the heterotetrameric subunit structure of the tuber enzyme. Part II involves stud… more

Production of bacterial cellulose from alternate feedstocks

Description: Production of bacterial cellulose by Acetobacter xylinum ATCC 10821 and 23770 in static cultures was tested from unamended food process effluents. Effluents included low- and high-solids potato effluents (LS and HS), cheese whey permeate (CW), and sugar beet raffinate (CSB). Strain 23770 produced 10% less cellulose from glucose than did 10821, and diverted more glucose to gluconate. Unamended HS, CW, and CSB were unsuitable for cellulose production by either strain, while LS was unsuitable for … more

Description: The long term aim of this project is to assess the feasibility of increasing the conversion of photosynthate into starch via manipulation of genes encoding enzymes that may be rate-limiting in starch biosynthesis. In developing storage tissues such as tubers, starch biosynthesis is regulated by the gene activation and expression of ADPglucose pyrophosphorylase, starch synthase, branching enzyme and other ancillary starch modifying enzymes, as well as the allosteric-controlled behavior of ADPglu… more

Enhancement of photoassimilate utilization by manipulation of the ADPglucose pyrophosphorylase gene. Progress report, [April 15, 1988--April 14, 1989]

Description: During this period researchers have been successful in determining the structure of the rice pyrophosphorylase gene. Potato tuber ADPglucose pyrophosphorylse purification and structure studies were carried out as well as recombinant DNA studies. Evidence suggests that the tuber form is made up of subunits with similar molecular weights and immunological relatedness. In contrast, the spinach leaf enzyme and presumably the maize endosperm species is composed of two dissimilar sununits encoded by … more
