Metabolic Engineering in Plants to Control Source/sink Relationship and Biomass Distribution

Description: Traditional methods like pruning and breeding have historically been used in crop production to divert photoassimilates to harvested organs, but molecular biotechnology is now poised to significantly increase yield by manipulating resource partitioning. It was hypothesized that metabolic engineering in targeted sink tissues can favor resource partitioning to increase harvest. Raffinose Family Oligosaccharides (RFOs) are naturally occurring oligosaccharides that are widespread in plants and are … more

The Stereochemistry of 1,4-Di-Tert-Butyl-1,4-Dihydronaphthalene

Description: A new approach was taken in the elucidation of the stereochemistry of the symmetrically 1,4-disubstituted 1,4-dihydronaphthalene systems which show deceptively simple NMR spectra. Thus, the stereochemistry of 1,4-di-tert-butyl-1,4-dihydronaphthalene was established unequivocally. The epoxide derivative, the subject compound, 1,4-di-tertbutyl-2, 3-epoxy-1, 4-dihydronaphthalene, was formed and its trans stereochemistry was established by ¹H NMR and ¹³C NMR. A lanthanide shift study was also condu… more

Homologs of Mammalian Lysosomal Lipase in Arabidopsis and Their Roles in Lipid Droplet Dynamics

Description: Lipid droplets (LDs) are organelles with many functions in cells and numerous protein interactors facilitate their biogenesis, maintenance, and turnover. The mammalian lipase responsible for LD turnover during lipophagy, LipA, has two candidate homologs in Arabidopsis: MPL1 and LIP1. One or both of these plant homologs may function in a similar manner to mammalian LipA, providing an LD breakdown pathway. To test this hypothesis, wild type (WT) Arabidopsis plants, MPL1 over-expressing (OE) muta… more

Tobacco Phospholipase D β1: Molecular Cloning and Biochemical Characterization

Description: Transgenic tobacco plants were developed containing a partial PLD clone in antisense orientation. The PLD isoform targeted by the insertion was identified. A PLD clone was isolated from a cDNA library using the partial PLD as a probe: Nt10B1 shares 92% identity with PLDβ1 from tomato but lacks the C2 domain. PCR analysis confirmed insertion of the antisense fragment into the plants: three introns distinguished the endogenous gene from the transgene. PLD activity was assayed in leaf homogenates … more

Structural Octatonicism in Cindy McTee's Symphony No.1: Ballet for Orchestra

Description: Cindy McTee's Symphony No. 1: Ballet for Orchestra is composed primarily of pitch materials from the octatonic collections that contain both diatonic and non-diatonic relationships in the themes, harmonic content, and larger structure of the symphony. Because the octatonic collections contain the potential for both diatonic and non-diatonic relationships, the piece is argued to have octatonic structure, as the octatonic collection is capable of producing both relationships. The second chapter c… more

Development and Testing of Fluorous Metal-Organic Frameworks (FMOFs) for Photoluminescent Sensing and Water Purification Applications

Description: In this work, fluorous metal organic framework 1 (FMOF-1) and its isomorphic structures are investigated for both sensing and filtration applications. In chapter 2 both FMOF-1 and its acetone derivative, aFMOF-1, are shown to exhibit an unusual photoluminescent behavior by having a "turn-on" emission when exposed to vacuum. This baroluminescence occurs at different rates depending on specific gas, allowing FMOF-1 and aFMOF-1 to not only act as vacuum gauges, but also potentially act as sensors … more

Effect of Amino Acids on Growth and Cartenogenesis in Corynebacterium Species Strain 7E1C

Description: Studies were evaluated on the effects of known growth factors on the growth and carotenogenesis of Corynebacterium species strain 7ElC. The complex medium, Tryptic Soy Broth, was found to stimulate growth and production of more pigment in the light and in the dark than did a mineral salts-glucose medium. A complete amino acid mixture added to LSG enhanced carotenogenesis in the dark in Corynebacterium 7ElC, while B-vitamins retarded carotenogenesis. No absolute requirement for one or more amino … more

Exploring the ROI of 1:1 Computing Programs at the High School Level

Description: This dissertation explored the cost and value of 1:1 computing programs in high schools in Texas. The study examines whether the total cost of ownership of these programs can be justified by the student testing gains and graduation rates. It investigates whether student learning outcomes show a definable correlation between positive gains and the implementation of 1:1 computing programs. The study also explores whether there is a measurable return on investment of 1:1 programs based on testing … more

Synthesis and characterization of 2-chloro-3-benzylthiopyrrolo[1,2-a]- benzimidazol-1-one and 2,3-di(benzylthio)pyrrolo[1,2-a]benzimidazol-1-one.

Description: The reaction between o-phenylenediamine and 2,3-dichloromaleic anhydride has been probed and found to give 2,3-dichloropyrrolo[1,2-a]- benzimidazol-1-one as the major product. Chlorine substitution in 2,3-dichloropyrrolo[1,2-a]benzimidazol-1-one by added benzylthiol occurs in the presence of pyridine to provide the corresponding monosulfide and disulfide derivatives. The first benzylthiol ligand undergoes reaction at the C-3 position of the five-membered pyrrolo-1-one ring, with the addition of… more

The Unpublished Works for Clarinet by Alexander Grechaninov: Preparing a Performance Edition of the Sonata No. 1 for Clarinet and Piano, Op. 161

Description: Alexander Grechaninov was one of the most important composers of the late Russian Romantic School. By the second half of the twentieth century he remained one of the few living composers who continued the traditions of the great Russian Romantic masters, such as Tchaikovsky and Rimsky-Korsakov. He is primarily known for his liturgical works, which are truly masterpieces of this genre. Because many of his instrumental works remain unpublished, particularly the chamber works, they continue to be … more

Specialized Lipid Droplet Associated Proteins are Required for the Selective Packaging of Various Lipid Classes into Lipid Droplets

Description: Simmondsia chinensis, better known as jojoba, is a desert shrub that is native to the southwestern United States and northern Mexico. Jojoba is unusual in that instead of primarily storing triacylglycerols (TAGs) in its seeds, it instead stores large quantities of highly valued liquid wax esters (WEs) as its major seed storage lipid. Unfavorable agronomical characteristics have unfortunately limited the production of these valuable lipids, and spurned efforts to produce these highly valued WEs … more
