Biological Rhythms: Implications for the Worker

Description: This report discusses biological rhythms: what they are, how they are controlled by the brain, and the role they play in regulating physiological and cognitive functions. The major focus of the report is the examination of the effects of nonstandard work hours on biological rhythms and how these effects can interact with other factors to affect the health, performance, and safety of workers.

Morning and Afternoon Response to Exercise Using Two Test Protocols

Description: The purpose was to investigate time of day on responses to two types of exhaustive cycle ergometer tests. Sixteen males performed six exercise tests: three - constant power protocol and three - incremental protocol. The first test was a learning trial; the other tests were performed one in the morning and one in the afternoon. ANOVA revealed that O2 deficit and lactate were higher in the afternoon than the morning. Regardless of test type, time to exhaustion tended to be higher in the afternoon. VO2max was unaffected by the time of day and test type. These results confirmprevious reports of a time of day effect on anaerobic capacity, and support the contention that V02max is stable about a day.

An Analysis of the Genesis of Motive, Rhythm, and Pitch in the First Movement of the Sonata for Two Pianos and Percussion by Béla Bartók.

Description: This dissertation presents evidence that Béla Bartók created his masterwork, the Sonata for Two Pianos and Percussion (1937), in a very complex period of his life. Since it was a mature piece, Bartók utilized typically "Bartókian" compositional techniques and styles. His ethnomusicological studies were also influential factors in the creation of the Sonata for Two Pianos and Percussion. We can be witness to how different the first draft was to the published version; the minor and major changes are revealed in the draft study of the Sonata for Two Pianos and Percussion 's first movement. These changes allow today's musicians to reconstruct the compositional process. The first movement introduces some interesting uses of sonata form, to be explored in more detail in the analysis. Starting with linear analysis, the basic motives and rhythmic patterns are discussed and supported with Bartók's own explanations. The conclusion of this study has important ramifications for performance: it eases up the pressure on the performers, since problematic passages are analyzed and explained - preparing the players' mentally for the performance. This is music which is hard to play and difficult to analyze. The analysis, combining the results of both theoretical and musicological studies, is intended to help both analysts and performers understand the genesis of the piece and, for performers, to execute the music in the best possible manner.

Heart rate and oxygen consumption during the critical prenatal period in chicken embryos (Gallus gallus): Influence of light cues and the onset of pulmonary ventilation.

Description: To examine if a rhythm can be entrained in either heart rate or oxygen consumption in late stage embryos (days 17-19.5) with light as a zeitgeber, chicken embryos were incubated in complete darkness (D:D) and 12:12 light:dark cycle (L:D). Light had no impact on oxygen consumption (390 µL O2∙min-1∙egg-1) but increased heart rate for non-internally pipped embryos (260 to 270 beats∙min-1 during light cycle). Oxygen consumption increased independent of pipping while heart rate increased (255 to 265 beats∙min-1) in D:D embryos due to pipping. A light-induced rhythm or effect occurred in heart rate but not oxygen consumption, suggesting heart rate and oxygen consumption may be uncoupled.

Final report

Description: The originally funded project was geared to pursue research on regulation of photosystem II (PSII) in the cyanobacterium Synechococcus elongatus PCC 7942. We characterized a locus, psfR, (psbA stimulating factor) that affects expression of the psbAI gene, which encodes the PSII protein D1. Over-expression of psfR, which encodes a protein with receiver and pseudo-receiver domains, acts at the promoter region to elevate expression of psbAI and a subset of other loci. We reoriented the remainder of the funding to make a greater impact through completion of a functional genomics project that had been initiated with funding from another agency. The goal is inactivation of each gene individually in the S. elongatus genome, and completion of the entire genome sequence. At the end of the project we will have screened all loci for involvement in circadian rhythms of gene expression and assembled an archived set of clones that can be used to create the mutations to screen for any other phenotype. During the project period we: (1) prepared a functional genomics website for S. elongatus PCC 7942 that posts sequences prior to GenBank release, and presents the strategy and progress for the genomics project (http://www.bio.tamu.edu/synecho/); (2) determined the sequence of and annotated the S. elongatus 46 kb plasmid, pANL; (3) submitted assembled sequences with annotation of 8 cosmid inserts to GenBank (313 kb), with sites of transposon insertions indicated; (4) mutagenized approximately an additional 600 kb of the genome (16 cosmids) and identified sequences flanking the mutations; (5) recombined mutagenesis substrates into the S. elongatus genome to produce gene inactivations (at the sites of transposon insertions) for approximately 415 kb of mutagenized sequence (85% of these have already been screened for circadian phenotypes) (6) identified the clpPIIclpX locus as important in determining circadian period; and (7) demonstrated effectiveness of antisense RNA ...
