Orlando Shooting Revives Debate over Restricting Blood Donations by Gay Men

Since the 1980s, the Food and Drug Administration (FDA) has recommended that men who have sex with men (MSM) be indefinitely deferred (i.e., permanently banned) from donating blood. Cuff-less Blood Pressure Measurement Using a Smart Phone

Description: Blood pressure is vital sign information that physicians often need as preliminary data for immediate intervention during emergency situations or for regular monitoring of people with cardiovascular diseases. Despite the availability of portable blood pressure meters in the market, they are not regularly carried by people, creating a need for an ultra-portable measurement platform or device that can be easily carried and used at all times. One such device is the smartphone which, according to comScore survey is used by 26.2% of the US adult population. the mass production of these phones with built-in sensors and high computation power has created numerous possibilities for application development in different domains including biomedical. Motivated by this capability and their extensive usage, this thesis focuses on developing a blood pressure measurement platform on smartphones. Specifically, I developed a blood pressure measurement system on a smart phone using the built-in camera and a customized external microphone. the system consists of first obtaining heart beats using the microphone and finger pulse with the camera, and finally calculating the blood pressure using the recorded data. I developed techniques for finding the best location for obtaining the data, making the system usable by all categories of people. the proposed system resulted in accuracies between 90-100%, when compared to traditional blood pressure meters. the second part of this thesis presents a new system for remote heart beat monitoring using the smart phone. with the proposed system, heart beats can be transferred live by patients and monitored by physicians remotely for diagnosis. the proposed blood pressure measurement and remote monitoring systems will be able to facilitate information acquisition and decision making by the 9-1-1 operators.

Effects of a methylcholanthrene-induced lymphosarcoma on the blood of DBA/1J and swiss white mice

Description: The investigation was concerned with characterizing effects of this tumor line on lipid metabolism in DBA/1J mice and serum protein levels and cellular changes in DBA/1J and Swiss white mice. Total lipids, lipid phosphorus, neutral lipids, and changes in fatty acids were determined in liver, spleen, skin and tumor of DBA/1J mice bearing the lymphosarcoma at various days after injection of tumor cells.

Responses to Diets High in Phenylalanine Compounds as Genetic Parameters in Mice

Description: The induction of phenylketonuria in mice through the use of excess dietary phenylalanine is an area in which limited research has been done. This study intends to pursue further work in this area, more specifically, to study the effects of excess dietary phenylalanine and the phenylalanine analogue A.P.B.A. (2-amino-3-phenyl butanoic acid) (7) on brain serotonin and brain norepinephrine. In addition, the effects of these two compounds on the incidence of audiogenic seizures will be explored.

Studies on Human Plasma Lecithin:Cholesterol Acyltransferase: Physical and Chemical Characterization and Coupled Spectrophotometric Enzyme Assay

Description: The physico-chemical properties of lecithin:cholesterol acyltransferase were investigated. The amino acid composition analysis showed a relatively high content of glutamic acid, aspartic acid, glycine and leucine. The spectrophotometric titration of phenolic groups in the enzyme showed a large increase in absorbance at 295 nm with an apparent pK of about 12.0. The largest change in molar ellipticity at 222 nm was also observed above pH 11. Circular dichroism studies revealed that human lecithin:cholesterol acyltransferase has a relatively high content of β-pleated sheet structure (48%) with 20% α-helix, and 32% remaining structure. Human lecithin:cholesterol acyltransferase has a high extinction coefficient at neutral pH. Microsequencing of the amino terminal residues of the enzyme revealed a hydrophobic character. Inactivation of lecithin:cholesterol acyltransferase activity was observed using diisopropylfluorophosphate with a stoichiometry of 1 mole of diisopropylphosphate incorporated per mole of enzyme. This suggests the involvement of a serine residue in the active site of the enzyme, possibly for the formation of an acyl-intermediate. A new quicker assay method for lecithin:cholesterol acyltransferase was developed. This assay involved coupling reaction with acyl CoA synthetase, ΡΡᵢ-dependent phosphofructokinase, aldolase, triosephosphate isomerase and α-glycerol-3-phosphate dehydrogenase monitoring a change in the absorbance or fluorescence intensity due to the oxidation of NADH. The activity of each coupling enzyme was accurately determined to establish the optimum assay condition for lecithin:cholesterol acyltransferase. The coupled enzyme assay for lecithin:cholesterol acyltransferase by spectrofluorometry showed a significant change in relative fluorescence intensity whereas a UV absorption spectroscopy method showed no significant absorbance change for the initial rate of lecithin:cholesterol acyltransferase reaction.

Evaluating the Pulse Sensor as a Low-Cost and Portable Measurement of Blood Pulse Waveform

Description: This study was aimed at determining whether the digital volume pulse waveform using the Pulse Sensor can be used to extract features related to arterial compliance. The Pulse Sensor, a low-cost photoplethysmograph, measures green light reflection in the finger and generates output, which is indicative of blood flow and can be read by the low-cost Arduino UNO™. The Pulse Sensor code was modified to increase the sampling frequency and to capture the data in a file, which is subsequently used for waveform analysis using programs written in the R system. Waveforms were obtained using the Pulse Sensor during two 30-s periods of seated rest, in each of 44 participants, who were between the ages of 20 and 80 years. For each cardiac cycle, the first four derivatives of the waveform were calculated and low-pass filtered by convolution before every differentiation step. The program was written to extract 19 features from the pulse waveform and its derivatives. These features were selected from those that have been reported to relate to the physiopathology of hemodynamics. Results indicate that subtle features of the pulse waveform can be calculated from the fourth derivative. Feature misidentification occurred in cases of saturation or low voltage and resulted in outliers; therefore, trimmed means of the features were calculated by automatically discarding the outliers. There was a high efficiency of extraction for most features. Significant relationships were found between several of the features and age, and systolic, diastolic, and mean arterial blood pressure, suggesting that these features might be employed to predict arterial compliance. Further improvements in experimental design could lead to a more detailed evaluation of the Pulse Sensor with respect to its capability to predict factors related to arterial compliance.
