Fuel gas production from animal waste: Phase I. Quarterly progress report (2nd), September 1, 1976--December 1, 1976. Dynatech report No. 1556

Description: During this second quarter, meetings were held with contractors on the ERDA Fuel Gas from Animal Waste Program to discuss project planning and review. Site visits were made to groups involved in anaerobic digestion. An engineering and economic analysis of a process for the anaerobic digestion of animal residue was completed; the results can be used for the design of a pilot plant unit. A comprehensive engineering report of this analysis was completed, and a first draft was submitted to ERDA. Se… more

Study of the effects of radiation of nucleic acids and related compounds. Progress report, August 15, 1975--August 14, 1976

Description: Ionizing radiation produces genetic effects in biological systems. Since genetic effects are usually the result of modifications of DNA or sometimes of RNA, interest is being centered on the chemical and physical nature of radiation-induced lesions to nucleic acids and their components. These investigations have revealed the enormous complexity of chemical events and the possible degradation of nucleic acids by strand breakage. Therefore, work in the ionization radiation of nucleic acids has pr… more

Energy and protein production from pulp mill wastes. Progress report, September 15, 1976--December 15, 1976

Description: Significant progress was made during the past quarter in establishing the operability and reliability of major pieces of equipment needed for the production of protein and methane from spent sulfite liquor (SSL). Batch ozonations of SSL were conducted at times varying from 1 to 6 hours at pH's in the range of 10 to 2. These screening experiments consistently indicate that low ph's favor the breakdown of SSL into organic fragments which are more easily assimulated by micro-organisms. Approximate… more

Technical progress report, August 1, 1975--July 31, 1976. [Biochemical studies on RNA of tobacco mosaic virus]

Description: Previous work had demonstrated the presence of a unique low-molecular-weight RNA component (LMC) in extracts of tobacco mosaic virus (TMV) infected tissue. Enough of this component has been isolated during the past year to ascertain that it has a molecular weight of 250,000 daltons and that it acts as an in vitro messenger for the synthesis of TMV capsid protein. Thus, we conclude that at least one monocistronic messenger RNA for a virion coded product is generated during TMV infection. Strains… more

Potential radiosensitizing antiviral and anticancer pyrimidine nucleosides. Technical progress report, June 1, 1975--December 22, 1975. [UV radiation]

Description: A newly developed method of synthesis now affords the large scale production of 5'-amino analogs of a variety of nucleosides. This procedure has been used to synthesize the 5'-amino analog of 5-iodo-, 5-bromo-, 5-chloro-, 5-fluoro-, 5-trifluoromethyl-2',5'-dideoxyuridine. In addition to the previously established antiviral activity of 5-iodo-5'-amino-2',5'-dideoxyuridine (AIU), we have found 5-bromo- (ABrU) and 5-iodo-5'-amino-2',5'-dideoxycytidine (AIC) to be even more potent antiviral agents … more

Effects of whole-body. gamma. -irradiation on the biosynthesis of certain serum proteins. Final report, November 29, 1967--June 30, 1976. [Rats]

Description: Whole-body exposure of rats to ionizing radiations yielded an increased incorporation of labeled amino acids into serum albumin in in vivo studies suggesting a stimulation of biosynthesis. Actually this may have been caused by an elevated hepatic transport of labeled amino acids (see below). A suppressed biosynthesis of albumin was observed when the experiments were performed in vitro using liver microsomes. Impaired biosynthesis appeared to be caused by a reduced mRNA production. Irradiation s… more

Cell-cycle-related biosynthesis. [Sarkosyl, neocarzinostatin, adriamycin]

Description: The state of chromatin during the cell cycle was examined using synchronized cultures of CHO hamster cells. Results support Mazia's dynamic chromosome cycle model and indicate that DNA-interactive chemotherapeutic agents elicit different types of kinetic responses in treated cells, suggesting a degree of specificity of interaction between various alkylating and intercalating agents and the genome. Effects of sarkosyl crystals, heparin, and chemotherapeutic agents, neocarzinostatin and adriamyci… more

DNA damage and repair mechanism. [DNA damage and repair mechanisms]

Description: The ability of cells to survive in an environment specifically damaging to its DNA can be attributed to a variety of inherent repair mechanisms. This is a form of repair in which alterations are directly reversed to their original form. This reversibility is exemplified by the photoreactivation of ultraviolet-induced pyrimidine dimers. This phenomenon is attributable to the action of an enzyme, photolyase (photoreactivating enzyme), which is able to monomerize the uv-induced pyrimidine dimers i… more

Comparison of the cytological effects produced by ionizing radiations of different LET. Progress report, April 1975--April 1976. [X radiation, fission neutrons, rats, eyes]

Description: Rat eyes were irradiated with a single dose of 1000, 850, 700, or 550 rads of x-radiation or 225, 200, 175, 150, or 125 rads of fission neutron radiation. Some eyes are being examined periodically for cataracts; others were tested by wounding the lens at 3, 14, or 28 days after irradiation. At 10 months after irradiation, the 1000-rad-X cataracts were a bit more advanced than the 225-rad-n group; this suggests an RBE of about 4. Wounding the lens initiated a ''wave'' of DNA synthesis followed b… more

Blood platelets and coagulation. Annual progress report, June 1, 1975--May 31, 1976. [/sup 75/Se tracer study of effects of thrombopoietin on platelet production in mice and rabbits]

Description: The primary emphasis during the past year has been extension of studies of platelet production and demonstration of a humoral agent (i.e., thrombopoietin) that regulates the production of platelets by megakaryocytes. Incorporation of selenomethionine-/sup 75/Se (/sup 75/SeM) into platelets was used to study the effect of partially purified fractions of plasma proteins upon platelet production in normal mice and rabbits. Significant increases in levels of /sup 75/SeM in circulating platelets wer… more

Biological conversion of biomass to methane. Quarterly progress report, September 1, 1976--November 30, 1976

Description: During the past quarter all experimental systems have become operational. The earlier problems with equipment acquisition have delayed the progress somewhat. However, most of these problems have been overcome such that the experimental programs have been initiated. This report will concentrate on the manure fermentation portion of this project. The study relating to the effect of reactor design is underway, but the results are too preliminary to evaluate. Therefore, no data from this activity w… more

Some Aspects of the Mechanism of Bacteriophage Function. Progress Report, February 11, 1975--January 31, 1976. [UV Radiation]

Description: Studies were conducted on replication, recombination, and maturation of E. coli phage lambda. Red-mediated dimers, earlier thought to be recombination intermediates, have been shown to involve extension of a single DNA rather than an interaction between two DNA's. Dimers have also been found in superinfected lysogens and may be an early stage of recombination. A recombination intermediate resembling a Holliday structure has been identified by electron microscopy. The lambda Red system has been … more

Enzymes and microorganisms in food industry waste processing and conversion to useful products: a review of the literature

Description: Bioconversion of food processing wastes is receiving increased attention with the realization that waste components represent an available and utilizable resource for conversion to useful products. Liquid wastes are characterized as dilute streams containing sugars, starches, proteins, and fats. Solid wastes are generally cellulosic, but may contain other biopolymers. The greatest potential for economic bioconversion is represented by processes to convert cellulose to glucose, glucose to alcoho… more
