Effector Response of the Aspartate Transcarbamoylase From Wild Type Pseudomonas Putida and a Mutant with 11 Amino Acids Deleted at the N-terminus of PyrB.

Description: Like its enteric counterpart, aspartate transcarbamoylase (ATCase) from Pseudomonas putida is a dodecamer of two different polypeptides. Unlike the enterics, the Pseudomonas ATCase lacks regulatory polypeptides but employs instead inactive dihydroorotases for an active dodecamer. Previous work showed that PyrB contains not only the active site but also the effector binding sites for ATP, UTP and CTP at its N-terminus. In this work, 11 amino acids were deleted from the N-terminus of PyrB and the ATCase with the truncated protein was expressed in E. coli pyrB- and purified. The wild type enzyme was similarly treated. Velocity-substrate plots without effectors gave Michaelis-Menten kinetics in all cases. Deleting 11 amino acids did not affect dodecameric assembly but altered effector responses. When carbamoylphosphate was varied, the mutant enzyme was inhibited by UTP while the wild type enzyme was activated 2-fold. When the aspartate was varied, CTP had no effect on the mutant enzyme but strongly inhibited the wild type enzyme.

Smoothing the information seeking path: Removing representational obstacles in the middle-school digital library.

Description: Middle school student's interaction within a digital library is explored. Issues of interface features used, obstacles encountered, search strategies and search techniques used, and representation obstacles are examined. A mechanism for evaluating user's descriptors is tested and effects of augmenting the system's resource descriptions with these descriptors on retrieval is explored. Transaction log data analysis (TLA) was used, with external corroborating achievement data provided by teachers. Analysis was conducted using quantitative and qualitative methods. Coding schemes for the failure analysis, search strategies and techniques analysis, as well as extent of match analysis between terms in student's questions and their search terms, and extent of match analysis between search terms and controlled vocabulary were developed. There are five chapters with twelve supporting appendixes. Chapter One presents an introduction to the problem and reviews the pilot study. Chapter Two presents the literature review and theoretical basis for the study. Chapter Three describes the research questions, hypotheses and methods. Chapter Four presents findings. Chapter Five presents a summary of the findings and their support of the hypotheses. Unanticipated findings, limitations, speculations, and areas of further research are indicated. Findings indicate that middle school users interact with the system in various sequences of patterns. User groups' interactions and scaffold use are influenced by the teacher's objectives for using the ADL. Users preferred to use single word searches over Boolean, phrase or natural language searches. Users tended to use a strategy of repeating the same exact search, instead of using the advanced scaffolds. A high percent of users attempted at least one search that included spelling or typographical errors, punctuation, or sequentially repeated searches. Search terms matched the DQ's in some instantiation 54% of all searches. Computer Supported Collaboration: Is the Transfer of Cognitive Structures Mediated by Mode of Communication?

Description: The objective of this study was to observe evidence of structural transfer among subjects in a group problem-solving activity and determine whether mode of collaborative technology or use of a priming agent affected the nature of transferred structures. Evidence for structural transfer is found in three theoretical perspectives: organizational ditransitive (linguistic) verb structures, adaptive structuration theory, and mental model transfer theory. Dependent variables included various grammatical structures and coefficients derived from pretest and posttest scores on David Kolb's Learning Styles Inventory, modified for the experiment. The combination of changes in grammatical frequencies and learning style may suggest that one or more media or the priming agent may affect structural transfer. Results indicate that groups using the GroupSystems™ collaborative technology produced less overall linguistic content than did subjects using a generic chat system, but employed more complex language as indicated by frequency of the organizational ditransitive verb structure. Also, subjects supplied with an organization chart (priming agent) during the group problem-solving session experienced greater change on the learning styles inventory than did those participating in the session without the chart. These findings suggest that mode of communication and use of priming agents may contribute positively or negatively to the transfer of structures among group members. Researchers, collaborative system designers, organizational leaders, trainers & educators, and frequent collaborative technology system end-users should be aware of these potential affects. Suggestions for future research are provided. Relationship of theoretical foundations of structural transfer to constructivism is discussed.

Establishing the utility of a classroom effectiveness index as a teacher accountability system.

Description: How to identify effective teachers who improve student achievement despite diverse student populations and school contexts is an ongoing discussion in public education. The need to show communities and parents how well teachers and schools improve student learning has led districts and states to seek a fair, equitable and valid measure of student growth using student achievement. This study investigated a two stage hierarchical model for estimating teacher effect on student achievement. This measure was entitled a Classroom Effectiveness Index (CEI). Consistency of this model over time, outlier influences in individual CEIs, variance among CEIs across four years, and correlations of second stage student residuals with first stage student residuals were analyzed. The statistical analysis used four years of student residual data from a state-mandated mathematics assessment (n=7086) and a state-mandated reading assessment (n=7572) aggregated by teacher. The study identified the following results. Four years of district grand slopes and grand intercepts were analyzed to show consistent results over time. Repeated measures analyses of grand slopes and intercepts in mathematics were statistically significant at the .01 level. Repeated measures analyses of grand slopes and intercepts in reading were not statistically significant. The analyses indicated consistent results over time for reading but not for mathematics. Data were analyzed to assess outlier effects. Nineteen statistically significant outliers in 15,378 student residuals were identified. However, the impact on individual teachers was extreme in eight of the 19 cases. Further study is indicated. Subsets of teachers in the same assignment at the same school for four consecutive years and for three consecutive years indicated CEIs were stable over time. There were no statistically significant differences in either mathematics or reading. The Effect of Average Grain Size on Polycrystalline Diamond Films

Description: The work function of hydrogen-terminated, polycrystalline diamond was studied using ultraviolet photoelectron spectroscopy. Polycrystalline diamond films were deposited onto molybdenum substrates by electrophoresis for grain sizes ranging from 0.3 to 108 microns. The work function and electron affinity were measured using 21.2 eV photons from a helium plasma source. The films were characterized by x-ray photoelectron spectroscopy to determine elemental composition and the sp2/sp3 carbon fraction. The percentage of (111) diamond was determined by x-ray diffraction, and scanning electron microscopy was performed to determine average grain size. The measured work function has a maximum of 5.1 eV at 0.3 microns, and decreases to 3.2 eV at approximately 4 microns. Then the work function increases with increasing grain size to 4.0 eV at 15 microns and then asymptotically approaches the 4.8 eV work function of single crystal diamond at 108 microns. These results are consistent with a 3-component model in which the work function is controlled by single-crystal (111) diamond at larger grain sizes, graphitic carbon at smaller grain sizes, and by the electron affinity for the intervening grain sizes.

Dimensions in Random Constructions.

Description: We consider random fractals generated by random recursive constructions, prove zero-one laws concerning their dimensions and find their packing and Minkowski dimensions. Also we investigate the packing measure in corresponding dimension. For a class of random distribution functions we prove that their packing and Hausdorff dimensions coincide.

College Freshman Biology Two Semester Course: Integrating Deep Processing Teaching Techniques

Description: Development of a college level freshman biology course was undertaken in response to government reports that American students have fallen behind students of other countries in the area of the sciences. Teaching strategies were investigated to accomplish two objectives, to define essential academic material to include in the course and to investigate teaching techniques that would increase deep processing of the information. An active process that consisted of applying the cognitive information to solving problems or developing answers to questions was defined as critical thinking. Critical thinking was incorporated into the course by the use of case studies.

The Federal Constitution and Race-Based Admissions Policies in Public Charter Schools

Description: The primary questions addressed in this dissertation are whether race-based admissions policies in charter schools are constitutionally permissible, and if not, how could an admissions policy be designed so that it would promote school diversity without violating the law? These questions are important because there are significant numbers of philosophers and scholars who hypothesize that student body diversity not only enhances educational outcomes but also is a necessary component of civic education in a liberal democracy. The researcher takes no particular stance on the benefits of educational diversity, focusing instead on the constitutional questions raised by the use of race-sensitive policies in the interest of diversity. The primary methodology used throughout is legal research, though the literature review includes references to political philosophers and social scientists as well as primary legal sources. Chapter I outlines the most frequent arguments made in favor of school diversity and suggests that the judicial philosophy expressed by the Supreme Court over the last twenty-five years has moved away from the philosophy expressed in Brown v. Board. In Chapter II, Supreme Court precedent on affirmative action policies is analyzed, focusing mainly on the decision of the divided Court in University of California Board of Regents v. Bakke. Chapter III provides a detailed analysis of how six different Federal Circuit Courts interpreted Bakke, highlighting numerous recurring judicial themes and concerns. In Chapter IV, existing charter school laws are examined state by state. Chapter V suggests several policy options for those interested in promoting a diverse charter school student body.

A Comparison of Texas Pre-service Teacher Education Programs in Art and the 1999 National Art Education Association's Standards for Art Teacher Preparation

Description: Texas programs in pre-service art teacher preparation vary little. Since 1970, the National Art Education Association (NAEA) has created voluntary standards in hopes of decreasing variability among programs. In 1999, the NAEA published Standards for Art Teacher Preparation, outlining 20 content areas that art pre-service programs should provide their students. To obtain information on the implementation and the extent to which these 20 standards are being implemented, a questionnaire was sent to all programs in Texas. The 20 standards were the dependent variable for the study. The four independent variables used in this ex post facto study were: the size of the institution where the program exists; the number of full-time art faculty; the number of full-time art education faculty; and, the number of undergraduate art education students who graduated last year. The 20 standards or provisions were scored on a Lickert scale with six options: zero (not taught) to five (comprehensively taught). The response size (N = 23) was 47% of the state's 49 approved programs. The results from the survey suggest no significant difference among programs. However, the results showed a significant difference in the number of provisions taught between programs with no art educators and those with 1 to 3 art educators. One art educator seemed to increase the number of pedagogical provisions taught but did not increase the extent or enhance the degree to which each provision was taught. A comprehensively taught response to the NAEA provisions on the questionnaire was further investigated through analysis of catalog course descriptions and correspondence with participants. The results are estimated in credit hours and indicate that there may be a point where time on task decides the limit that constitutes a comprehensive preparation. The Effects of Selective Serotonin Reuptake Inhibitors (SSRI) on Auditory Measures in Women

Description: This study examined the relationship between selective serotonin reuptake inhibitor (SSRI) medication and auditory measures in clinically depressed women. Experimental subjects were tested in both a medicated and unmedicated condition. Experimental subjects were compared to a normal control group; additionally intrasubject comparison was made within the experimental group. Test measures included: audiometry, tympanometry, otoacoustic emissions, uncomfortable loudness level, masking level difference, SCAN-A, Synthetic Sentence Identification (SSI), and the low predictability section of the Revised Speech in noise (RSPIN). The unmedicated group scored significantly less favorably than the control group on the following tests; SCAN-A (composite, filtered words, and auditory figure ground), R-SPIN (0MCR condition in both the right and left ears). Additionally, the unmedicated group scored significantly less favorably than the medicated group on the SSI (-20MCR condition right ear only) and of the R-SPIN (0MCR condition right ear only). Other test measures indicated consistent trends but did reach significance.

Hypotheses for Scratch Behavior of Polymer Systems that Recover

Description: Scratch recovery is a desirable property of many polymer systems. The reason why some materials have demonstrated excellent scratch recovery while others do not has been a mystery. Explaining the scratch resistance based upon the hardness of a material or its crosslink density is incorrect. In this thesis, novel polymers were tested in an attempt to discover materials that show excellent scratch recovery - one of the most important parameters in determining the wear of a material. Several hypotheses were developed in an attempt to give an accurate picture of how the chemical structure of a polymer affects its scratch recovery. The results show that high scratch recovery is a complex phenomenon not solely dependent upon the presence of electronegative atoms such as fluorine.

Carl Gottlieb Reissiger (1798-1859) Forgotten Composer for the Clarinet

Description: Carl Gottlieb Reissiger was a successful German composer, conductor, and teacher in the first half of the nineteenth century. At the height of his career, he was Hofkapellmeister of theater and opera in Dresden, a position he held until his death. He was a composer of more than 200 works in a multitude of different genres. Today he is mainly known as a composer of opera, a small portion of his total output as a composer. He wrote approximately eighty piano solos, eighty collections of songs or duets, nine masses, and many smaller sacred choral works, as well as 27 piano trios, seven piano quartets, and three piano quintets. In addition to these many works, he wrote five works for the clarinet: Concertino, op. 63, Duo Brillant for clarinet and piano, op. 130, Fantasie, op. 146, Second Fantasie, op. 180, and Adagio und Rondo alla polacca, op. 214. This document provides a biographical sketch of Reissiger, including his personal life, his life as a conductor, and his life as a composer. It also provides a look at the artistic life of his day: his fellow composers and the music they were writing for clarinet, outstanding clarinetists and the different instruments they were playing. The aim of this study is to provide a stylistic analysis of Reissiger's five works for clarinet, including a discussion of form, melody, harmony, and rhythm. This document puts forth the proposal that these works are worth resurrecting and that Reissiger, as a composer of clarinet music, is more than just a secondary composer.

Sonata for Piano (1963) by Sergei Michailovich Slonimsky: Musical Analysis and Discussion on Interpretation and Performance

Description: The essay begins with the overview of Russian-Soviet piano music from the second half of the 19th century to the end of the 20th century. Then, biographical information about Sergei Slonimsky and an overview of his major compositions is provided. The majority of the paper focuses on Slonimsky's Sonata for Piano (1963). A brief discussion of the Sonata's compositional history is followed by the formal analysis of the overall structure of the work. Slonimsky's original principle of organization of the music is emphasized: the system of constant interrelation of the main thematic material combined with elements of the sonata-allegro form. In the analysis of the harmonic language of the piece, the composer's extensive use of Russian folk elements such as diatonic melodies, sigh motives, parallel triads, and simultaneous use of the lower third with the major triad is pointed out. The rest of the paper focuses on issues of interpretation and performance. Special notice is given to the problem of incorporating a percussive type of playing with the elements of folk cantilena singing. The paper concludes with the history of Sonata's performances and a discussion of current recordings.

General Process in the Creation of Estruendos and Principal Structural Elements of the Composition

Description: My composition, Estruendos, is a work for large symphonic orchestra, guitar and computer-generated and processed sounds on CD. The work lasts 23 minutes and 45 seconds. My dissertation is composed of two parts: Part One comprises the analysis and Part Two comprises the score. Part One gives a brief background of my compositional dialect and aesthetics. It also includes a discussion of the compositional process and general overview of Estruendos. In addition, it illustrates the primary role the placement of sonic events in time and timbral structure play in the pathos of Estruendos.

The Effects of Selective Serotonin Reuptake Inhibitors (Ssri) on Auditory Measures in Clinically Depressed Subjects.

Description: The purpose of this study was to investigate the relationship between selective serotonin reuptake inhibitor (SSRI) medication on auditory skills in clinically depressed subjects. Experimental subjects prescribed an SSRI were tested in a medicated and an unmedicated condition, and the test results were compared. Furthermore, the experimental group was compared with a control group consisting of normal subjects. Test measures included pure tone audiometry, tympanometry, acoustic reflex thresholds, and auditory electrophysiologic measures such as auditory brainstem and auditory late responses. An assessment scale for depression (Beck Depression Inventory-II) was also used. Results indicated statistically significant differences for the BDI-II between the control and experimental groups for both conditions. Electrophysiologic measures indicated a significantly shorter latency for auditory late potential P1 at 55 dBnSL, and a significantly larger amplitude at 45 dBnSL for the N1/P2 component for the unmedicated group. Although the other measures showed trends, they did not reach significance.

La Primavera: Concertino for English Horn and Chamber Orchestra

Description: La Primavera: Concertino for English Horn and Chamber Orchestra is a work in a traditional chamber orchestra instrumentation: single woodwinds (flute, oboe, clarinet, and bassoon), two French horns, trumpet, timpani and strings. A through-composed work of 14 minutes in duration, the Concertino is conceptually based on the idea that spring is not the first of the seasons, but rather the last. As a result, all of its motivic materials are organically linked to one another, and function as paired forces that struggle for supremacy. The introduction of the third motive functions as a motivic synthesis, since it contains intrinsic elements of previous motives. There are several important compositions based on the topic of the seasons among them we find: Vivaldi's Concerto Grosso Le Quatro Staggione, Haydn's oratorio The Seasons, and Piazzola's chamber work Las Estaciones. While researching this topic, the conceptual dilemma of spring as the last season was considered. This became a turning point in the compositional process strong enough to consider the spring as a singular topic of interest. The analysis of this work through Derrida's Deconstruction theory first came to me while reading Rose Rosengerd Subotnick's Deconstructive Variations: Music and Reason in Western Society. The Linguistic approach, was inspired in part by Leonard Bernstein's lecture “The Unanswered Question,” and Jean J. Nattiez's Music and Discourse: Toward a Semiology of Music.

Competency to Stand Trial: A Systematic Evaluation and Validation of the GCCT, MacCAT-CA, and ECST as Competency Measures

Description: Competency to stand trial cases constitute the largest percentage of forensic referrals for clinical psychologists. Furthermore, research suggests that the use of forensic measures facilitates the decisions of competency made by forensic examiners. This study investigated the construct validity of three competency measures: (a) the GCCT-MSH, (b) the MacCAT-CA, and (c) the ECST with 100 adult males incarcerated at the Tarrant County Jail in Fort Worth, TX. Construct validity was investigated via the use of a multitrait-multimethod research design for the three-prong conceptualization of the Dusky standard. Results indicated that current competency measures do an adequate job of assessing for factual understanding, but lack construct validity for two prongs: rational understanding and the ability to consult with counsel. In addition, the atypical presentation scales of the both GCCT and the ECST performed well at screening individuals for feigning. Finally, prediction of competency from clinical variables was also investigated. Psychotic symptoms and overall impairment were the strongest predictors of incompetency.

A Correlative Study of Gender and Social Style.

Description: This study examines the concepts of social style and gender to determine if a relationship exists between the two constructs. The hypotheses suggested a direct relationship between the categories of the BSRI (masculine, feminine, androgynous, and undifferentiated) and the Social Style Analysis (driver, amiable, expressive, and analytical). Ninety-four participants completed two self-report surveys. Chi-square analysis performed on the data found a significant relationship between feminine and amiable as well as androgynous and expressive. While the analysis suggested that masculine/driver and undifferentiated/analytical were not independent, the relationship found was not significant.

Synthesis of Crown Ether/Ammonium Salt for Electron Transfer Study

Description: The theoretical model of Beratan and Onuchic predicts a large attenuation of ET rates through hydrogen bonds; however, the effect of individual hydrogen bond on electron transfer reaction has not been systematically studied. The organic complexes in this study are a series of crown ether/ammonium salt, which incorporate a redox partner on each component of the complex. The dimethoxynaphthalene redox donor was attached to the crown ether and a series of ammonium salts was synthesized which bear substituted quinone and naphthoquinone acceptor. The complexes characterization and preliminary electron transfer rate measurement were completed with UV/Vis and steady-state emission spectroscopy.

A Measurement System for Monitoring Play in Typically Developing Children and Children with Autism

Description: A comprehensive measurement system was developed to monitor play in children with autism and typically developing children. The study was conducted in a preschool operated in conjunction with a center-based program for children with autism. The development of the measurement system was based on observations of four children with autism and three typically developing children during social and play activites. Data were collected on material use and several dimensions of play: Simple Manipulation, Functional Manipulation, Symbolic Toy Play, Symbolic Role Play and Play Themes. The results indicated that the measurement system consistently measured a wide range of play behaviors across children and materials. Significance of the information gathered from the measurement system in assessing play and designing interventions is discussed.

The Use of Genetic Polymorphisms and Discriminant Analysis in Evaluating Genetic Polymorphisms as a Predictor of Population

Description: Discriminant analysis is a procedure for identifying the relationships between qualitative criterion variables and quantitative predictor variables. Data bases of genetic polymorphisms are currently available that group such polymorphisms by ethnic origin or nationality. Such information could be useful to entities that base financial determinations upon predictions of disease or to medical researchers who wish to target prevention and treatment to population groups. While the use of genetic information to make such determinations is unlawful in states and confidentiality and privacy concerns abound, methods for human “redlining” may occur. Thus, it is necessary to investigate the efficacy of the relationship of certain genetic information to ethnicity to determine if a statistical analysis can provide information concerning such relationship. The use of the statistical technique of discriminant analysis provides a tool for examining such relationship.

An investigation of the current status of fund raising activities and training within student affairs divisions in Texas colleges and universities.

Description: The primary focus of this study was to discover the depth of involvement with fundraising by student affairs professionals in Texas. It sought to determine the predominance of chief student affairs officers trained in development and the types of training that they received. Cooperation between student affairs divisions and development offices was also studied and whether there was a correlation between a cooperative relationship and the number of successful fundraising goals. This study includes a review of related literature on student affairs fundraising, a description of the methodology, results of the survey, conclusions, implications, and recommendations that may assist in future decision-making concerning future involvement in fundraising. The surveys were mailed to 149 four-year (public and private) institutions and two-year public institutions in Texas. The senior staff members of both the student affairs office and development office were asked to complete a survey. There was a 60.7% return rate consisting of responses from 72 development offices and 95 student affairs offices for a total of 167 usable responses. The study found that 59% of the student affairs officers had some formal training and/or on the job training. Involvement in fundraising was reported by 62.1% of the chief student affairs officers. Eighteen percent reported that they employed a development officer exclusively for student affairs fundraising, and another 30% had a development officer assigned to student affairs. Most development officers and student affairs officers perceived the other officer as cooperative rather than competitive in raising funds. Recommendations from this study include studying community college fundraising structures separately for more depth, conducting qualitative interviews with student affairs development officers, making a comparison of student affairs offices that have full-time development officers, and comparing the differences in fundraising success between development officers and chief student affairs officers. The Effects Of The Allocation Of Attention Congruent With Lateralized Cognitive Tasks On EEG Coherence Measurements

Description: The single task condition of the Urbanczyk and Kennelly (1991) study was conducted while recording a continuous electroencephalographic (EEG) record. Attention was allocated by instructed lateral head orienting and eye gaze either congruently or incongruently with lateralized cognitive tasks. Thirty university subjects retained a digit span or a spatial location span for a 20 second retention interval. EEG data were extracted from the 20 second retention intervals and interhemispheric coherence was calculated for homologous sites in the temporal, parietal and occipital regions of the brain. There was a main effect for group, with congruent orienting producing greater coherence values than incongruent orienting. This effect of attention on alpha coherence values was found in the low alpha (8-10 Hz) frequency band. This provides evidence that the lower alpha frequency band is reflective of manipulations of attention. The higher coherence measures for the congruent orienting group indicates that homologous regions of the two hemispheres are more coupled into a single system when lateralized attention activates the same hemisphere performing the cognitive task. In the higher alpha frequency band (11-13 Hz) group, sex, site and task interacted. This provides evidence that the higher alpha band is more affected by cognitive processing of the specific task undertaken. An interhemispheric brain system, affected by the lateral orientation of attention, may underlie psychometric intelligence's general “g” ability (Spearman, 1927.)

The Effects of Non-differential Reinforcement and Differential Reinforcement on Problem Behaviors and Accuracy of Responding of Autistic Children.

Description: The effects of non-differential reinforcement and differential reinforcement on problem behaviors and accuracy of responding of autistic children was examined. In experiment 1, one child with autism participated, and in experiment 2, two children with autism participated. In the non-differential reinforcement condition both prompted and unprompted responses were reinforced. In the differential reinforcement condition only unprompted responses were reinforced. Overall, problem behaviors were more frequent in the non-differential reinforcement condition. In experiment 1, accuracy was higher in the differential reinforcement condition, while experiment 2 showed inconclusive results with regards to accuracy. It is concluded that non-differential reinforcement can decrease problem behaviors in teaching situations, but may not be sufficient to ensure acquisition of target tasks.
