Adenylate Energy Charge Determinations of Soil Bacteria Grown in Soil Extract Medium

Description: The adenylate energy charge values of twenty bacteria isolated from soil and cultured in a medium consisting of soil and distilled water were determined by the luciferin-luciferase bioluminescense method. The purpose of this study was to examine the growth and energy charge values of these organisms in soil extract medium, and to determine what effect the addition of glucose has on their energy charge values. Three of the organisms employed in this study showed energy charge values similar to t… more

Advanced Techniques in Microbial and Molecular Biology: Laboratory Procedures for a Graduate Level Course

Description: Advanced laboratory techniques for Microbial and Molecular Biology at the graduate level are presented in this thesis. The procedures for the laboratory experiments are set forth in detail. This laboratory is conducted as two parts, each by a different professor. Part 1 covers the experiments conducted by Dr G.A.O. Donovan. These experiments include an introduction, staining procedures, biochemical reactions, mutagenesis experiment, essays,. preparation and analysis of plasmid DNA and various o… more

An Analysis of Distribution Patterns of Amphibians and Reptiles in Texas

Description: The purpose of this investigation was to analyze the distribution of amphibians and reptiles in Texas by means of the methods of Webb and Hagmeier and Stults. An additional graphical analysis was made, including range and range limits which provides a cross-section of faunal change along selected base lines across the state.

Application of Fluorescent Antibody Methods for the Enumeration and Identification of Bacillus Cereus

Description: This particular work is proposed as a test of the expedience of using the fluorescent-antibody technique as a method for enumeration and identification of certain strains of B. cereus that have been found to be effective in preventing taste and odor in water supplies resulting from certain Actinomycete blooms.

Applications of Remote Sensing and GIS to Modeling Fire for Vegetative Restoration in Northern Arizona

Description: An accurate fire model is a useful tool in predicting the behavior of a prescribed fire. Simulation of fire requires an extensive amount of data and can be accomplished best using GIS applications. This paper demonstrates integrative procedures of using of ArcGIS™, ERDAS Imagine™, GPS, and FARSITE© to predict prescribed fire behavior on the Kaibab-Paiute Reservation. ArcGIS was used to create a database incorporating all variables into a common spatial reference system and format for the FARSIT… more

Assessment of Changes in Aquatic Macrophyte Occurrence Following Introduction of Triploid Grass Carp in a North Texas Reservoir

Description: The objectives of this project were to measure changes in frequency of occurrence of submerged macrophytes over the first two growing seasons following stocking with triploid grass carp at two fish per acre and to measure differences in macrophyte biomass between areas excluded from herbivory and adjacent control sites after 16 months following establishment of exclosures. The project also seeks to measure concentrations of fluridone following the herbicide treatment, and to compare two methods… more

Assimilation of Organic Carbon by Aquatic Actinomycetes

Description: There were two purposes of this investigation: the first was to develop a method for studying spore germination, hyphal growth and mycelial development of the aquatic actinomycetes under the microscope so that the life cycle of this organism could be continuously followed. The second purpose was to determine the rate of carbon utilization from various types of nutritional sources, and to correlate this with the colony development and spore formation.

Biological Control of the Red Imported Fire Ant by the Entomopathogenic Nematode, Steinernema Carpocapsae (Weiser)

Description: Field trials were conducted in 1988 to evaluate the effectiveness of Steinernema (=Neoaplectana) carpocapsae (Weiser) in controlling the fire ant. Infective juveniles (IJ) of the nematode were applied as drench on 235 and 422 mounds, respectively for 2-month summer and 6-week fall evaluation periods. In comparative trials, amidinohydrazone (Amdro) was applied to 249 (summer) and 65 (fall) active mounds, with 245 (summer) and 78 (fall) untreated active as controls. Nematode treatments resulted i… more

Biological Indices of Stream Pollution

Description: A thorough biological survey and evaluation is a lengthy and expensive project. The number of technically trained persons required prevent its use by most public agencies. Since public health departments are the groups most frequently concerned with measuring the effects of pollution in streams, a need exists for a simplified method of sanitary survey. Recognizing this need, the Texas Health Department assigned the writer the problem of devising a method of survey which would require a field pa… more

Characterizing Spontaneous Neurophysiological Activity with Measures of Statistical Serial Dependence: a Summary Statistic and an Extension of the Joint Interval Histogram

Description: Two measures which indicate statistical serial dependence were evaluated. The n-dimensional Average Stored Information index (ndASI) is a measure of conditional information, which compares the entropies of higher order conditional distributions to estimate average statistical serial dependence. The generalized ranked joint interval histogram (RJIH-o) is a new nonparametric graphical analysis method. It extends the joint interval histogram by depicting longer interval sequences and can be interp… more

Chemical Composition of the Peptidoglycan of Vitreoscilla Stercoraria

Description: The peptidoglycan layer of Vitreoscilla stercoraria, ATCC 15218, was isolated from intact cells after treatment with sodium lauryl sulfate (SLS) and digestion with Pronase. Amino acid and amino sugar content was analyzed and 67% of the total present was made up of glutamic acid, alanine, diaminopimelic acid (DAP), and glucosamine in a molar ratio of 1:1.7:1:0.7. Electron microscopy of the final peptidoglycan product showed a thin, delicately folded sacculus which exhibited a morphology differen… more
