Actinomycetes are able to utilize a great variety of carbohydrates, like sugar. The particular kind of sugar and its concentration has decisive effect on the growth of microorganisms. The proper nutritional media aids also in the production of spores. Based on this generalization, that the growth and sporulation of microorganisms are greatly influenced by the nature and the concentration of carbohydrates, an attempt has been made to study Actinoplanes philippinensis with respect to this influence.
Advanced Molecular and Microbial Techniques: a Complete Laboratory Notebook
The purpose of this project is to produce a complete and thorough notebook that may be used to supplement laboratory coursework. Its intent is to be used primarily by the students to aid them in understanding background information and the proper laboratory procedures involved in various types of experiments. The laboratory notebook is a summation of all the experiments and procedures used in the six-credit hour Advanced Microbial and Molecular Biology (BIOL 5160) course offered during the summer semester at the University of North Texas. This class is a team taught effort by Professors O'Donovan and Kunz. The course is constructed as an intensive practice exercise to teach the student about gene mutations, biosynthetic pathways, preparation and analysis of plasmid DNA, and many other topics included in the notebook.
Anatomical and Morphological Responses of Cardiospermum Halicacabum L. (Balloon Vine), to Four Levels of Water Availability
C. halicacabum (Sapindaceae) is an invasive plant that is considered a nuisance species in Texas riparian environments. Little is known of the tolerance of C. halicacabum to flooding and drought; however, this information may provide insight into the characteristics that contribute to C. halicacabum purported invasiveness. C. halicacabum seedlings (n = 92) were exposed to one of four levels of water availability (flooded, saturated, intermediate and dry) over six weeks under greenhouse conditions. Plant performance was affected by water availability; however, there was no effect on survivorship. Flooded and saturated plants exhibited morphological adaptations; producing adventitious roots, hypertrophy, and aerenchyma tissue. Morphological measures, anatomical responses, and patterns of biomass allocation all indicate that C. halicacabum is able to survive periodic inundation, perform in saturation, and establish and thrive on the drier end of a moisture gradient.
Anatomy and Physiology Syllabus for Community Colleges
This syllabus includes both lecture notes and laboratory activities for a two-semester anatomy and physiology community college course. The syllabus is based on a 16-week semester that is comprised of a three-hour lecture section and a one-hour laboratory class each week. Both the lecture course and laboratory are necessary to fulfill the requirement for anatomy and physiology. Laboratory activities coincide with lectures to enhance understanding of each topic by providing visual and hands-on experiments for the concepts learned in the lecture. Laboratory quizzes will be given each week to help students maintain a working knowledge of the material learned in the laboratory. This course is appropriate for the typical anatomy and physiology student and should benefit both students who plan to major in biology and also those who are non-biology majors. Because subject matter in anatomy and physiology is quite difficult, the importance of attendance and good study skills is stressed.
Aquatic Vegetation Nutrient Budgets and Sedimentation in a Southwestern Reservoir
During four growing seasons, aquatic vascular plant production and distribution were studied in Pat Mayse Lake, Texas, a 2425 hectare oligo-mesotrophic reservoir. The dominant macrophyte population was Myriophyllum spicatum L. Growth rates and regrowth rates of mechanically harvested Myriophyllum beds were found to be dissimilar. Based on estimates of watermilfoil nutrient content, there were insufficient nutrients in the entire population to alter the trophic status of this reservoir should all of the nutrients be instantaneously released. Sediments were the primary nutrient (nitrogen and phosphorus) sink. Bank erosion and solids transport from the watershed appear to contribute most of the sediments and a lake-wide mean sedimentation rate of 2.5 cm/year was estimated from sediment trap and core sample data.
An Assay Method for Determining Extra-Cellular Lipases from Pseudomonas aeruginosa
The applicability of an isotopically labelled assay system to determine the lipase production in Pseudomonas aeruginosa was evaluated. Supernatant from cultures of Pseudomonas aeruginosa grown in a medium containing olive oil was incubated with a substrate containing labelled trioleate. Fatty acids were isolated by means of a liquid-liquid partition system. Enzyme activity was determined by measuring the amounts of free fatty acid by liquid scintillation counting. Findings indicate that the isotopicallylabelled, liquid-liquid partitioning assay is reliable, sensitive and adaptable to rapid assay conditions. It was also determined that different strains of Pseudomonas aeruginosa produce varying amounts of lipase. Partial purification of supernatant by gel filtration produced two protein peaks showing enzymatic activity.
Assessing Effects of an Environmental Education Field Science Program Fostering Responsibility at an Urban Middle School
The study investigated the ability of an extracurricular program to influence environmental responsibility of sixth and seventh graders. The Children's Environmental Attitude and Knowledge Survey (CHEAKS) was evaluated for appropriateness in assessing the worth of this particular environmental education strategy emphasizing water quality fieldwork and technology. CHEAKS is designed with psychometric reliability and validity that may be used in comparing disparate programs. Wilcoxon two sample tests were used to analyze data gathered from two student groups; one participated in an "Enviro-Mentals Club"; the other received no treatment. Analysis showed no significant change in environmental attitudes between groups, but did show significance (p <= 0.05) in environmental knowledge growth. Therefore, the investigated program had marginal success in influencing environmental responsibility.
Bacterial Challenge in Lumbricus Terrestris: A Terrestrial Invertebrate Immunotoxicity Model.
A bacterial challenge assay was developed utilizing the earthworm, Lumbricus terrestris, in order to assess potential immunotoxic effects from exposure to specific polychlorinated biphenyl congeners. Earthworms were inoculated with Aeromonous hydrophila, establishing a 10-day LD50. In vitro assays for effects of PCBs on phagocytosis agreed with mammalian studies, demonstrating potent suppression of phagocytosis by the non-coplanar PCB congener 138 and no suppression by the coplanar congener 126. However, when the effects of the two PCB congeners were evaluated for suppression of resistance to a whole animal infection challenge assay, coplanar PCB 126 decreased the ability of L. terrestris to withstand infection while non-coplanar PCB 138 did not.

Bioconcentration and Morphological Effects of Triclosan on Three Species of Wetland Plants
Triclosan (TCS) is an antimicrobial compound found in several types of common household products. After being washed down the drain, TCS will then end up in the local watershed. Although numerous studies have been conducted to evaluate the fate and effects of TCS in aquatic environments, there have been no studies evaluating the role arbuscular mycorrhizal fungi (AM) play in a plants response to TCS exposure. Three species of wetland plants native North Texas were inoculated with AM spores and exposed to 0, 0.4 g/L and 4.0 g/L TCS concentrations. Root morphology of E. prostrata and S. herbacea showed AM and exposure responses. S. herbacea produced the greatest amounts biomass and TCS bioaccumulation, in all but one treatment. It also displayed opposing results to E. prostrata in measures of root length, root surface area, relative root mass, relative shoot mass and shoot:root ratio. TCS root tissue concentrations increased with increased exposures for both E. prostrata and S. herbacea. Even though E. prostrata had the lowest levels in each measure of biomass production, it had the highest amount of root TCS bioaccumulation in the AM inoculated 4.0 g/L treatment. H. laevis was between the other two species in terms of biomass production, and did not demonstrate any exposure or inoculation effects in morphology or TCS accumulation. Overall, no clear patterns were detected, which highlights the fact that further study is required to completely understand the effects compounds like TCS can have on plant community structure, and ultimately ecosystem function.

Changes in Heterozygosity Through Time in American Standardbred and American Saddlebred Horses (1960-1990)
Observed and expected heterozygosity (H) levels for ten electrophoretic blood marker loci and expected H for seven red blood cell (RBC) anitgen/antibody loci were examined for 20 years in American Standardbred and 30 years in American Saddlebred horses. Standardbreds were classed by gait, Trotter and Pacer, and evaluated separately in most analyses. 4,404 Trotters and 12,271 Pacers were found to have statistically highly significant losses of mean total observed H through time for the ten electrophoretic loci (P<0.005), although in Trotters the loss was more extreme (P<<0.001). Loss of H in 5984 Saddlebreds was not significant (P=0.259). Correlations of RBC expected H through time showed decreases in all three groups.
Characterization of the OCT Plasmid-Encoded Mercury Resistance Genetic Locus in Pseudomonas putida
A 17.1 Kb genetic element encoding for mercury resistance (OCT-Hg^r) was shown to translocate from its original location on the OCT plasmid to the resistance plasmid, RPl, in Pseudomonas putida. Analysis of RPl-Hg^r recombinant plasmids revealed that insertion of mercury resistance genes into RPl could occur at a variety of sites, with all recombinants having common EcoRI restriction fragments of 9.4, 3.8, 2.3, and 1.6 Kb, derived from the insertion. Hybridization analysis suggested the existence of extensive homology between this insertion and the prototypic mercury resistance transposon, Tn501, as well as the location of a similar merA sequence. Although the overall size was shown to be quite different from Tn501, striking physical similarities are shared between these two elements.
Circulatory and Respiratory Responses to Cycle Ergometry at Different Pedal Rates
The effects of moderate workload exercise at different pedal rates on circulatory and respiratory parameters were studied. Five subjects performed seven discontinuous constant-load cycle ergometer tests of 30 minutes duration at pedal rates of 40, 50, 60, 70, 80, 90 and 100 rpm. Oxygen uptake and carbon dioxide production were determined by standard open circuit spirometry, while heart rate was recorded by electrocardiograph. The COâ‚‚ rebreathing procedure was administered during the exercise bout in order to determine cardiac output. Blood pressure was determined for each test, and total peripheral resistance was calculated. The findings indicate that progressive increases in pedal frequency during discontinuous constant-load cycle ergometry produce progressive increases in cardiovascular, respiratory and metabolic responses and a decrease in gross exercise mechanical efficiency. The results indicate that oxygen uptake, cardiac output, heart rate, ventilation and arterial-venous oxygen difference increases curvilinearly as pedal rate increases, possibly as a result of increases in recruitment of muscle fibers and/or muscle groups. These findings suggest that circulatory and respiratory responses are due to "central command" which sets the basic efferent response pattern. However, this effector pattern is modulated by afferent input originating from the legs during progressive increases in pedal rate.
College Freshman Biology Two Semester Course: Integrating Deep Processing Teaching Techniques
Development of a college level freshman biology course was undertaken in response to government reports that American students have fallen behind students of other countries in the area of the sciences. Teaching strategies were investigated to accomplish two objectives, to define essential academic material to include in the course and to investigate teaching techniques that would increase deep processing of the information. An active process that consisted of applying the cognitive information to solving problems or developing answers to questions was defined as critical thinking. Critical thinking was incorporated into the course by the use of case studies.
Combined Effects of Polycyclic Aromatic Hydrocarbons and Ultraviolet Light on Benthic and Pelagic Macroinvertebrates
Crude oil commonly enters freshwater aquatic ecosystems as thin sheens forming on the water surface. Oil contains mixtures of toxic compounds called polycyclic aromatic hydrocarbons (PAHs), some of which are known to be photodynamic, increasing toxicity when combined with ultraviolet radiation. Benthic macroinvertebrate communities are commonly utilized as bioindicators, and as such rely on abundant data in literature concerning benthic macroinvertebrates' relative tolerances to a wide range of pollutants. A series of 10 plastic traps, half of which were filtered from UV radiation, were deployed in an urban pond for 27 days to determine colonization preferences of benthic macroinvertebrates to UV exposure. Results of this in situ experiment indicated that the majority of aquatic insects collected from traps inhabited the UV exposed treatment group, particularly the nonbiting midge, Chironomidae. A series of bioassays were then completed to investigate the sensitivities of a Chironomidae species to thin sheens of crude oil in the presence and absence of UV radiation. All bioassays were conducted using 10 day old Chironomus dilutus larvae cultured in the lab. The series of C. dilutus bioassays were all conducted under the same water quality parameters, temperatures, and oil sheen dosing methods, under a 16:8 photoperiod and exposed to 16 h UV per day. Five replicates (n = 20) were loaded into 350 mL glass crystallizing dishes and exposed to four treatments for 96 hours: no UV/with sheen, with UV/no sheen, both UV and oil sheen, and a control. Three assays with 175 mL water volume were completed, one with no sediment, one with silica sand, and one with fine sand. Sediment type had a significant effect on mortality (p < 0.0001), but significant effects of UV or PAHs were not found. Comparative Toxicity of Refuse-Derived Fuel Fly Ash on Two Species of Earthworms, Lumbricus terrestris and E. foetida, Using an Artificial Soil Exposure Protocol
Research estimated toxicity of refuse-derived fuel fly ash (RDF-FA) on two earthworms species, Lumbricus terrestris and Eisenia foetida. Specific objectives were to: (1) Compare their 14-day LC50s under light and dark conditions; (2) separate toxicity due to osmotic, pH and physical factors from that of heavy metal contaminants; (3) compare relative differences of artificial soil and commercial soil as exposure media for evaluating toxicity to earthworms. The 14-d LC50s for L. terrestris in dark and light were 57.0 and 48.34 % RDF-FA, and 59.25 and 41.00 % RDF-FA for E. foetida using artificial soil. All of the toxicity resulted from heavy metals within the RDF-FA. Using L. terrestris, the LC50s for artificial soil and commercial soil were 52.30 and 64.34%.
Comparison of Heteranthera Dubia (Jacq.) MacM.-associated Macroinvertebrates Between Georgraphical Regions in the United States
Macroinvertebrates associated with the aquatic plant, water stargrass (Heteranthera dubia), were sampled from 12 waterbodies in four regions of the United States from June to August 2005. Taxa richness, evenness, and diversity were lowest in the Lower Midwest (LMW) region, and higher in Northern sites, especially the Upper Midwest (UMW), and Northeast (NE). While relative abundance varied from site to site and region to region, utilization of the plant by functional groups remained fairly constant. Collector-gatherers consistently comprised the largest portion of invertebrates sampled. The shredder/ herbivore functional group comprised an average of 17 % of total groups. Through an exhaustive literature review, it was found that shredder/ herbivores of water stargrass have not been reported in the literature. Because of this, the herbivore group was analyzed separately and consisted of 2,383 specimens representing 23 species. The most common groups were Rhopalosiphum sp., Nectopsyche spp. and chironomids. No differences were found in herbivore diversity or evenness between sampling regions, but species richness was significantly different.
A Comparison of Predicted and Actual Trophic Status of Lake Ray Roberts, Texas Based on Chlorophyll A
Two years before impoundment, the trophic status of Lake Ray Roberts was predicted by applying the total phosphorus input into the Organization for Economic Cooperation and Development (OECD) eutrophication model. Predicted mean summer epilimnetic (MSE) chlorophyll a of Elm Fork arm, Isle duBois arm and Main Body were in the eutrophic category of the OECD model. Observed MSE chlorophyll a two years after impoundment of Elm Fork arm, Isle duBois and Main Body had not reached their predicted means and were at the mesotrophic-eutrophic boundary of the OECD model. Six years after impoundment, observed MSE chlorophyll a for Main Body, was closer to its predicted mean and in the eutrophic category of the OECD model. Six years after impoundment, Elm Fork arm was the most productive area of Lake Ray Roberts. Observed means of chlorophyll a, total phosphates, suspended solids and turbidity were often highest in the Elm Fork arm. Wastewater effluent from Gainesville and Valley View, TX, had an impact on productivity in Elm Fork arm.
Comparison of Remediation Methods in Different Hydrogeologic Settings Using Bioplume II
A contaminant fate and transport computer model, Bioplume II, which allows simulation of bioremediation in ground water systems, was used to compare the effects of 11 remediation scenarios on a benzene plume. The plume was created in three different hydrogeologic settings from the simulation of an underground gasoline storage tank leak.

Conservation, Connectivity, and Coexistence: Understanding Corridor Efficacy in Fragmented Landscapes
Conservation corridors, areas of land connecting patches of natural land cover, are frequently cited and implemented as a restorative strategy to counteract fragmentation. Current corridor ecology focuses on experimental corridor systems or designed and built conservation corridors to assess functionality. Such systems and designs are typically short, straight swaths of homogenous land cover with unambiguous transitions between patches. Quantifying the degree to which amorphous landscape configurations, tortuosity, and heterogeneity of land cover and land uses within the corridor has on functional connectedness is a crucial yet overlooked component of corridor efficacy studies. Corridor literature lacks a robust and repeatable methodology for delineating existing landscape elements, recognizing arbitrary edges, and identifying the start and end of ambiguous transitions between the patches and corridor. Using a set of landscapes being studied as part of a global assessment of corridor efficacy, I designed a workflow that standardizes the boundary of corridor-patch interfaces. The proposed method is a quantitative and repeatable approach that minimizes the subjectivity in corridor delineations. This research investigates the degree to which the existence of a corridor modifies the structural and functional connectivity between patches connected by a corridor compared to an intact reference area.
Construction of a Physical Map of Moraxella (Branhamella) catarrhalis Strain ATCC25238
In order to gain a better understanding of this microorganismand its role in human pathogenesis, a physical map of Moraxella catarrhalis type strain ATCC25238 was constructed using pulsed field gel electrophoresis (PFGE) in combination with Southern hybridization techniques. Restriction endonucleases Not I, Rsr II, and Sma I were used to digest the chromosomal DNA. An overlapping circular map was generated by cross-hybridization of isolated radiolabeled fragments of Moraxella catarrhalis genomic DNA to dried PFGE gels. The number and location of the 16S and 23S ribosomal RNA genes were determined by digestion with l-Ceul enzyme and by Southern hybridization. Virulence-associated genes, the gene for β-lactamase, and housekeeping genes were also placed onto the physical map.
Cytotoxicity and Functional Toxicity of Mefloquine and the Search for Protective Compounds
Mefloquine hydrochloride is an antimalarial agent that has been used for the past 40 years. Numerous reports of neurological side effects have recently led the FDA to issue a strong warning regarding long-term neurological effects. This warning lead to the U.S. Army’s Special Forces and other components to discontinue its use in July of 2013. Despite reported adverse side effects, mefloquine remains in circulation and is recommended to travelers going to specific Asian countries. Mefloquine has been used as a treatment for those already infected with the malaria parasite (blood concentrations ranging from 2.1 to 23 µM), and as prophylaxis (blood concentrations averaging 3.8 µM) (Dow 2003). The purpose of this study was to quantify Mefloquine’s toxicity using spontaneously active nerve cell networks growing on microelectrode arrays in vitro and to identify compounds that alleviate or reduce toxic effects. The current literature on mefloquine toxicity is lacking electrophysiological data. These data will contribute to research on the mechanism of adverse side effects associated with mefloquine use. Sequential titration experiments were performed by adding increasing concentrations of mefloquine solution to cultured neurons. Network responses were quantified and reversibility was examined. In each network, activity decreases were normalized as a percent of reference activity yielding a mean IC50 value of 5.97 ± 0.44 (SD) µM (n=6). After total activity loss, no activity was recovered with two successive medium changes. To test for network response desensitization resulting from sequential applications over 5-6 hr periods, one-point titrations at varying concentrations were conducted with fresh networks. These experiments yielded a single concentration response curve with an IC50 value of 2.97 µM. This represents a statistically significant shift (p < 0.0001) to lower concentrations of mefloquine, demonstrating that sequential applications result in network desensitization. Detection of Mercury Among Avian Trophic Levels at Caddo Lake and Lake Lewisville, TX
Mercury (Hg) is a globally distributed toxicant that has been shown to have negative effects on birds. in the United States, avian taxa have been shown to possess high Hg concentrations in the northeast, Great Lakes and Everglades ecosystems; however, few studies have measured avian Hg concentrations in other geographic regions. Previous studies have documented high Hg concentrations in multiple organisms in east Texas, but birds were not included in these studies. the main objective of the present study was to quantify Hg concentrations in birds in differing trophic levels at Caddo Lake and Lake Lewisville, TX. Results suggest that Hg concentrations may be high enough to negatively impact some bird taxa, particularly those at high trophic levels, residing at both Caddo Lake and Lake Lewisville.
Development and Application of an Assessment Protocol for Watershed Based Biomonitoring
With numerous bioassessment methodologies available, a regional protocol needs to be developed to ensure that results are comparable. A regional assessment protocol was developed that includes collecting five benthic macroinvertebrate samples, identifying organisms to genus, and calculating the following metrics: Number of Taxa, Total Number of Individuals, Simpson's Diversity Index, Shannon's Diversity Index, Percent Contribution of Dominant Taxa, Hilsenhoffs Biotic Index, and Percent Contribution of Dipterans. Once the protocol was developed, it was used to assess the Bayou Chico tributaries and watershed. All three tributaries had been significantly impacted by human activity as had the watershed as a whole. This study indicates that a regional protocol could be developed and is appropriate for biomonitoring at the watershed scale.
Development and Validation of a Ramping Treadmill Protocol for the On-Line Measurement of Four Aerobic Parameters
Previously, Whipp et. al. (J. Appl. Physiol.: Respirat. Environ. Exerc. Physiol. 50(1):217-221, 1981) demonstrated the feasibility of determining four parameters of aerobic function, identified as maximum oxygen uptake (μVO_2), VO_2 at anaerobic threshold (θan), the time constant for oxygen uptake kinetics (rVO_2) and work efficiency (η), using a short duration ramped bicycle ergometer exercise test. Because of the importance of being able to measure these parameters on a variety of measurement instruments a short duration ramping treadmill protocol has been developed. The ability of this protocol to determine the four aerobic parameters has been validated against conventional methods. The results of this investigation indicate that μVO_2, θan, rVO_2 and, η may be obtained from a single, short-duration ramping treadmill test.
Developmental Patterns of Metabolism and Hematology in the Late Stage Chicken Embryo (Gallus Domesticus) at Two Incubation Temperatures.
How temperature affects physiological development in the chicken embryo is unknown. Embryos incubated at 38°C or 35°C showed no difference in growth or survival. The time to hatching was longer in 35°C than 38°C embryos (23.7 vs. 20.6 days), but unaffected was the relative timing of appearance of developmental landmarks (internal, external pipping). At stage 43-44, 38°C embryos maintained oxygen consumption around 1 mL/g/h despite acute temperature reduction (suggesting thermoregulatory maturation), unlike 35°C embryos. In 35°C embryos the lower oxygen-carrying capacity and temperature insensitive blood O2 affinity (P50 about 30 mmHg) may restrict O2 delivery to tissues, limiting metabolism during decreased ambient temperature. Reduced incubation temperature retards normal hematological and thermoregulatory development.
The Distribution of Pathogenic Pseudomonas aeruginosa in Sewage
The purpose of this study was to extend our understanding of the ecological relationships of P. aeruginosa by investigating the differences or similarities between the strains of this organism found in sewage and those found as pathogens in human infections. This research was approached by comparing the serological types of P. aeruginosa isolated from sewage contaminated waters in Argentina (South America) to those isolated from sewage contaminated waters in Texas. They were typed with sera obtained using P. aeruginosa isolated from human infections. The data obtained revealed that bacteria isolated from sewage in Texas and from soil and water in Argentina are antigenically similar to those isolated from human infections.
Distribution of Phosphates in a Sewage Plant and its Receiving Waters
The purpose of this paper is to detail the distribution and quantity of soluble phosphates in an activated sludge plant, its discharge creek, and receiving reservoir. The resultant analyses and comparison of data should provide some better understanding of the fate of phosphorus in a Southwestern waterway system.
Early Function of a Virulent Staphylococcal Phage
Early function of a temperature-sensitive mutant of staphylophage 44A HJD was examined during the twenty-five-minute period following infection. Host cell and phage DNA were labeled with C and3H respectively. DNA was separated into linear and covalently closed circular (CCC) forms by density-gradient centrifugation. The host, S. aureus, shows no CCC DNA, and apparently carries no plasmid. Following infection with wild type phage, CCC DNA forms occur in tritiated and 1 C DNA fractions 10 to 15 min after infection. Infection with mutant at permissive temperature also demonstrates CCC DNA with both labels. Infection with mutant at nonpermissive temperature produced no CCC DNA during the first 25 min after infection. The impaired function in this mutant may be a linker protein.
Effect of Amino Acids on Growth and Cartenogenesis in Corynebacterium Species Strain 7E1C
Studies were evaluated on the effects of known growth factors on the growth and carotenogenesis of Corynebacterium species strain 7ElC. The complex medium, Tryptic Soy Broth,was found to stimulate growth and production of more pigment in the light and in the dark than did a mineral salts-glucose medium. A complete amino acid mixture added to LSG enhanced carotenogenesis in the dark in Corynebacterium 7ElC, while B-vitamins retarded carotenogenesis. No absolute requirement for one or more amino acids was found,indicating a multiple amino acid requirement. The fewest amino acids found to stimulate carotenogenesis in the dark were a combination of those in the Serine and Histidine families which include serine, glycine, cysteine, and histidine.
The Effect of Human Alpha Interferon on Rat Kidney Cell Infected with Temperature-Sensitive Mutant of Rous Sarcoma Virus
LA31-NRK and B77-NRK are established cell lines that were normal rat kidney cells transformed with temperature-sensitive mutant (LA31) and wild-type Bratislava 77 (B77) of Rous sarcoma virus. It is recognized that many transformation-induced changes differentiate between normal and transformed cells. Morphology and four parameters of transformed cells such as saturation density, anchorage independence, plasminogen activator, and colony stimulating factor were used as indicators to observe the effect of human alpha interferon on the growth of NRK, LA31-NRK and B77-NRK. The results show that interferon could neither reverse the transformed cells to normal fashion nor change their behaviors or cause release of protease.
Effect of Indole-3-Acetic Acid on the Nucleic Acids of Synchronous Cultures of Chlorella Pyrenoidosa
It was the purpose of this study to investigate the effect of various concentrations of IAA on the nucleic acids of Chlorella pyrenoidosa TX 7-11-05. The time during the life cycle when the greatest effect occurred was investigated by the use of synchronous cultures.
Effect of Inorganic Phosphate on the Morphology and Poly-β-Hydroxybutyrate Content of Azotobacter Vinelandii Atcc 12837
This study was designed to determine the effect of inorganic phosphate in the growth media of A. vinelandii on the synthesis and accumulation of poly-β-hydroxybutyrate (PHB) and cellular morphology. Burk's media with no added phosphate and ten times normal phosphate resulted in altered cells, lower levels of protein, and decreased cell populations. PHB accumulation was much higher in the cultures with no added phosphate; however, the polymer granules differed in appearance from those of normal cells and had a decreased rate of utilization. The amount of PHB deposited in cells grown in media containing ten times the normal phosphate level was lower than that found in regular media.
The Effect of Light on Carotenoid Synthesis in Corynebacterium 7E1C
The effects of light, light "mimicking" chemicals, and protein synthesis inhibitors on the photo-induced carotenogenesis of Corynebacterium 7EIC were studied. Changes in the dosage of fluorescent light applied to dark grown cells showed a dose related carotenogenic response. Maintaining the same dosage but varying the wavelength of monochromatic light revealed that light with a wavelength of 280 to 450nm was responsible for photo-induction. It further showed a peak of photo-induction between the wavelengths of 370 and 430nm. The light "mimicking" chemicals antimycin A and p-Chloromercurybenzoate were shown to have no light "mimicking" effects. The transcriptional inhibitor of protein synthesis actinomycin D partially inhibited, and chloramphenicol a translational inhibitor, completely inhibited photo-induced carotenogenesis.
The Effect of Nine Diet and Water Combinations on the Culture Health of Ceriodaphnia Dubia
Culture health of Ceriodaphnia dubia was evaluated for organisms cultured using all combinations of three foods and three waters. Criteria used to assess health of cultures included adult and neonate weights, time required to produce first broods, neonate production, adult survival, and resistance to hexavalent chromium. Diet/water combinations which produced the most neonates were not found to produce adults which were more resistant to chromium than those which produced fewer neonates. Of those evaluated, a diet of Selenastrum capricornutum and a yeast-trout chow-cereal leaf mixture was best for culturing and testing Ceriodaphnia. The best synthetic water tested was a mixture of nine parts reconstituted hard water and one part bottled mineral water.
Effect of pH on the Persistence and Toxicity of Cyfluthrin to Chironomus Tentans
The effect of pH upon the aquatic toxicity of cyfluthrin was determined in 48 h static acute toxicity tests using 2nd instar Chironomus tentans larvae. Tests were conducted in both dechlorinated tap water and lake water of pH 8.0, 7.2, and 6.6. After 48 h, immobilized and dead larvae were removed and replaced with 2nd instar larvae to assess the persistence of toxicity. Midges were cultured in water adjusted to the pH values used in testing. Toxicity if cyfluthrin varied inversely with pH. An increase in the pH of tap water by 2 units resulted in a 2-decrease in toxicity. Toxicity of cyfluthrin also varied between tap and lake water of the same pH. EC50 values in lake water were approximately 2-3 times lower than those in tap water. Toxicity in the lake water and tap water at every pH tested was also significantly different when regression line elevations were compared. Natural waters amended with cyfluthrin were consistently more toxi to the chironomids than tap water of the same pH. Persistence of cyfluthrin at low pH also influenced chironomid behavior. Recovery of normal behavior generally began within 24 h at pH 8.0 At pH 6.0, recovery did not begin until one week after dosing. The persistence of cyfluthrin also varied with pH. Averaged across all concentrations, 30% of the initial dose remained in tap water (pH 8.0) after 48 h, compared with 45% (pH 7.2), and 75 % (pH 6.6).
Effector Response of the Aspartate Transcarbamoylase From Wild Type Pseudomonas Putida and a Mutant with 11 Amino Acids Deleted at the N-terminus of PyrB.
Like its enteric counterpart, aspartate transcarbamoylase (ATCase) from Pseudomonas putida is a dodecamer of two different polypeptides. Unlike the enterics, the Pseudomonas ATCase lacks regulatory polypeptides but employs instead inactive dihydroorotases for an active dodecamer. Previous work showed that PyrB contains not only the active site but also the effector binding sites for ATP, UTP and CTP at its N-terminus. In this work, 11 amino acids were deleted from the N-terminus of PyrB and the ATCase with the truncated protein was expressed in E. coli pyrB- and purified. The wild type enzyme was similarly treated. Velocity-substrate plots without effectors gave Michaelis-Menten kinetics in all cases. Deleting 11 amino acids did not affect dodecameric assembly but altered effector responses. When carbamoylphosphate was varied, the mutant enzyme was inhibited by UTP while the wild type enzyme was activated 2-fold. When the aspartate was varied, CTP had no effect on the mutant enzyme but strongly inhibited the wild type enzyme.
Effects of an Acute Bout of Near-Maximal Intensity Exercise on the Cardiac Enzymes in Human Sera
The Cardiac Profile, a pattern of serum enzyme changes seen within seventy-two hours after an AMI, is diagnostic aid for detecting occurrence of infarcts. The effects of exercise stress on the Cardiac Profile aid clinicians in avoiding diagnostic errors in patients immediately after exercise. Five male volunteers ran from six to ten miles. Serum enzyme levels were monitored serially three days before and five days after stress. Enzyme activity was determined spectrophotometrically and electrophoretically. Significant increases in total CPK and LDH were seen. An LDH 'one-two flip' occurred eight hours after exercise. No MB-CPK was found following the run.
Effects of Daily Oral Injections of Quercetin on Implanted Colon-25 Tumor Growth in Balb-C Mice
The effects of three oral dosages (0.4 mg, 0.8 mg, and 1.6 mg) of quercetin on Colon-25 tumors implanted in Balb-c mice were studied. The data in this study show that: (1) certain dosages of quercetin in alcohol solutions, reduces the weight, and size of implanted Colon-25 tumors in Balb-c mice, (2) these same dosages of quercetin all produce a profound neutrophilia combined with a significant lymphopenia at day 20 post-implantation, and (3) there was relatively little evidence of histological changes in the quercetin-treated tumor section which would indicate that the action(s) of quercetin is primarily at the subcellular level probably within the nuclei of the tumor cells.
The Effects of Hexadecanol on the Microbiota of Lake Hefner
It seemed desirable to investigate more fully the effect of hexadecanol on the microbial population of a reservoir. It was the purpose of this investigation to determine the effect of hexadecanol on the micro-biota of Lake Hefner, to ascertain which organisms were stimulated by hexadecanol both in the laboratory and the reservoir, and to investigate the degradation of hexadecanol by microorganisms selected from Lake Hefner.
Effects of Lysolecithin on Macrophages
The effect of lysolecithin on the macrophage was studied using five macrophage function assays. The results of indicate that lyso lecithin is a macrophage activating agent which causes enhanced cell spreading, increased phagocytosis of sheep erythrocytes, heightened membrane activity in the presence of damaged autologous red blood cells, chemotaxis, and vigorous phagocytosis and intracellular killing of Staphylococcus albus.
The Effects of Probiotics on Growth, and Metabolism in Juvenile Oreochromis mossambicus (Mozambique Tilapia)
Improving growth, lowering mortality rates, and having a faster turnaround to harvest is essential for the future of commercial aquaculture. The primary goal of this study was to determine if introducing a single strain probiotic Lactobacillus rhamnosus IMC 501 into the feed regimen of a commercially important aquaculture freshwater fish, Mozambique tilapia (Oreochromis mossambicus), would decrease mortality; change metabolic rates; and increase tissue wet mass (MW), standard length, growth rate and feed conversion rate (FCRs). IMC501 was added to the fishmeal in four increasing concentrations and compared to a control without probiotics. Results from two-way ANOVAs showed that both treatment levels and elapsed time had a significant effect on both mean standard length and wet mass; in the latter case, time points and treatments interacted with one another, showing that tilapia grew best with a moderate level of probiotics present. The growth benefits of probiotics continued for months after the initial treatments. Oxygen consumption (metabolic rate) was measured using closed respirometry and resulted in recording the first values for juvenile tilapia treated with probiotics. For oxygen consumption, there were significant treatment and time effects with significant interactions, indicating that metabolism increased with probiotics once the dosage exceeded three times the industry level. These results are consistent with the observed increases in mass, length and growth rates. These results demonstrate the importance of conducting dose-response experiments in order to determine the most effective concentration of probiotics in juvenile freshwater fish. Importantly, probiotics at the right concentration increase metabolic rates and can positively influence tilapia growth, which is of interest for the optimization of Mozambique tilapia production in aquaculture.

The purpose of this paper is to report the results of studies on the aerobic, mesophilic intestinal flora of guinea pigs subjected to conditions similar to those encountered by man in spacecraft.
Effects of Turbidity on Gilling Rates and Oxygen Consumption on Green Sunfish, Lepomis cyanellus
Laboratory studies conducted at 5, 15, 25, and 35 C measured changes in gilling rates and oxygen consumption of green sunfish in response to exposure to bentonite clay suspensions. The tests indicate that gilling rates are not affected by bentonite clay suspensions below 2125 FTU at 5 C, 1012 FTU at 15 C, and 898 FTU at 25 C. At turbidity levels exceeding 1012 FTU at 15 C and 898 at 25 C, gilling rates increased 50-70%. Tests were inconclusive at 35 C. Oxygen consumption rates were found to be unaffected by turbid suspensions below 3500 FTU at all four temperatures. Evidence suggests that increased gilling rates under highly turbid conditions are a means of compensating for reduced respiratory efficiency and a strategy for maintaining a constant oxygen uptake. Evidence indicates that the cost of increased gilling rates is probably met by a reduction in activity.
Evaluating Fish Impingement and Entrainment at the Comanche Peak Steam Electric Station
This study was designed to determine if impingement and entrainment by cooling water intake at the Comanche Peak Steam Electric Station have an adverse impact upon the Squaw Creek Reservoir fish population. The yearly impingement of fish was estimated to be 262,994 of 14 species. The threadfin shad (Dorosoma petenense) accounted for 96% of this total. Entrainment of eggs and larvae for a five month period was estimated to be 15,989,987 and 42,448,794 respectively. Two fish population studies were performed on Squaw Creek Reservoir to help assess impact. It was determined that the losses due to impingement and entrainment have no adverse impact upon the fish population of Squaw Creek Reservoir.
Evaluating the Habitat Requirements of the Golden Orb Mussel (Quadrula Aurea) for Conservation Purposes
Many freshwater mussels are imperiled, due to a number of interrelated factors such as habitat alteration, degradation of water quality, and impoundments. The Golden Orb mussel (Quadrula aurea, I. Lea, 1859) is endemic to the state of Texas and is currently a candidate for the endangered species list, as the number of known populations has been declining in recent years. Little is currently known about Q. aurea aside from basic distribution data. This study is focused on evaluating a combination of macro-habitat and micro-habitat variables to determine their influence on the distribution and density of this species. Macro-habitat variables, including dominant land cover, surface geology, and soil erodibility factor, did not have a significant relationship with mussel distributions. The best model of micro-habitat variables that impacts the Q. aurea distributions is comprised of relative substrate stability (RSS) at moderate flows and current velocity at low flows. For all mussel species in this study, current velocity at low flows is the primary variable that influences distribution. Q. aurea are associated with habitats where larger sediment particles (large gravel and cobble) help to stabilize the substrate in areas with higher current velocities. An understanding of the preferred habitats for Q. aurea can be used to help focus conservation efforts and practices.
Evaluation of a Constructed Wetland to Reduce Toxicity from Diazinon at the Pecan Creek Wastewater Treatment Plant, Denton, TX
The City of Denton Pecan Creek Wastewater Treatment Facility has periodically failed effluent toxicity testing. A Toxicity Identification Evaluation has determined that Diazinon in the effluent is contributing to the observed toxicity. Chlorpyrifos is also implicated as a factor. The City of Denton constructed a half acre experimental wetland to remove Diazinon related toxicity. Results from spiking and microcosm experiments indicate that the wetland can reduce the Diazinon.
An Evaluation of the Short-Term Embryo-Larval and Seven-Day Larval Test Methods for Estimating Chronic Toxicity of Zinc to the Fathead Minnow (Pimephales promelas)
Chronic toxicity of zinc to Pimephales promelas was estimated by conducting replicate static and static-renewal short-term embryo-larval tests and static-renewal seven-day larval tests. The two test methods were highly reproducible. Daily renewal of test solutions had little effect on the toxicity of zinc, however, the stage of development at which exposure was initiated affected the sensitivity of the toxic endpoints measured. The most sensitive and reproducible endpoint in the embryo-larval tests was survival of viable (non-deformed) larvae and in the seven-day larval test was growth of the larvae, which was slightly more sensitive than the embryo-larval test endpoint. The estimated MATC of 0.18 and 0.15 mg/L mean total and mean soluble zinc, respectively, compared well with published results. Because of its advantages and similar sensitivity, the short-term embryo-larval test was recommended for estimating chronic toxicity.
