Synthesis of high specific activity (1- sup 3 H) farnesyl pyrophosphate

Description: The synthesis of tritiated farnesyl pyrophosphate with high specific activity is reported. trans-trans Farnesol was oxidized to the corresponding aldehyde followed by reduction with lithium aluminium tritide (5%-{sup 3}H) to give trans-trans (1-{sup 3}H)farnesol. The specific radioactivity of the alcohol was determined from its triphenylsilane derivative, prepared under very mild conditions. The tritiated alcohol was phosphorylated by initial conversion to an allylic halide, and subsequent trea… more

Charge separation in photoredox reactions

Description: This report deals with the photoreduction of alkylmethylviologens in vesicles in which the combined effect of the alkylmethylviologen chain length and the addition of cholesterol to the vesicle system has been assessed. Although the alkyl chain length effect predominately controls the photoreduction yield it is shown that cholesterol addition can tune the magnitude of this effect. A series of neutral alkylphenothiazines has been synthesized and the effect of alkyl chain length in variously char… more

Cell-mediated mutagenesis and cell transformation by chemical carcinogens

Description: Results are reported from studies that showed that mutagenesis of mammalian cells can be achieved by carcinogenic polycyclic hydrocarbons, nitrosamines, and aflatoxins when tested in the presence of fibroblasts and hepatocytes which are able to metabolize these carcinogens. Further, we have found that there is a relationship between the degree of mutant induction and the degree of carcinogenicity of the different chemicals tested. By simultaneously measuring the frequency of cell transformation… more

Nuclear medicine technology. Progress report for quarter ending September 30, 1979

Description: Adrenal uptake in rats of /sup 117m/Sn-labeled 23-(trimethyl stanna)-24-nor-5..cap alpha..-cholan-3..beta..-o1 is demonstrated. The adrenal:blood and adrenal:liver ratios were 33:1 and 9:1, only one day after intravenous administration. A variety of /sup 117m/Sn-labeled steroids with structural modifications were also prepared to determine their effects on the adrenal uptake of these unusual steroids. Structural features required for significant adrenal uptake were determined. The diffusion cha… more

Biochemical endpoints of glucocorticoid hormone action

Description: Both the rapidly evolving metabolic effects of glucocorticoids and the more slowly developing lethal actions appear to be initiated via the synthesis of new mRNAs and proteins. The chronic suppression of cell growth may be the consequence of suppression of overall rates of protein synthesis (and probably RNA and DNA synthesis as well) that in turn may represent the cellular response to the small changes in ratios of adenine nucleotides that result from the suppression of oxidative ATP productio… more

Nuclear medicine technology progress report for quarter ending June 30, 1979

Description: Diffusion chamber assay system studies on the relative cytotoxicity and target cell uptake of /sup 195m/Pt-labeled cis- and trans-dichlorodiammineplatinum(II) (cis- and trans-DDP) are reported. These two agents are excellent candidates for such studies since cis-DDP is an established potent antitumor agent while the trans-isomer exhibits little antitumor activity. The KB tumor cell population in chambers from the cis-DDP-treated animals decreased while the cell population in chambers from the t… more

Description: The preparation of a unique class of trimethyltin-substituted steroids is described that were designed because of the potential use of the /sup 117m/Sn-labeled analogs for adrenal imaging. The trimethyltin-substituted steroids were readily prepared on the microscale in yields ranging from 40 to 80%. Studies with the /sup 123m/Te-labeled fatty acids have continued, and radiolabeled 6-telluraheptadecanoic acid and 11-telluraheptadecanoic acid have shown pronounced heart uptake in rats. Efficient microscale preparation of tin-117m-tin tetrachloride-a pivotal intermediate for the synthesis of tin-117m-labeled radiopharmaceuticals

Description: A microscale technique has been developed for the preparation of /sup 117m/SnCl/sub 4/. The radiolabeled tin tetrachloride is an important intermediate for the synthesis of /sup 117m/Sn-labeled organic radiopharmaceuticals. The flow-through system involves high temperature chlorination of metallic /sup 117m/Sn with subsequent cryogenic collection of the /sup 117m/SnCl/sub 4/ in a special reaction vessel. This vessel can then be used for the conversion of the tin tetrachloride to various useful … more

Bile Salt Surfactants in Micellar Electrokinetic Capillary Chromatography: Application to Hydrophobic Molecule Separations

Description: Bile Salt surfactants are used in the micellar electrokinetic capillary chromatography (MECC) separation of various hydrophobic compounds. The use of methanol in the mobile phase allows the separation of previously intractable compounds including polyaromatic hydrocarbons. The effects of methanol on critical micelle concentration is investigated for sodium dodecyl sulfate (SDS) and the bile salt sodium cholate. It is determined that the unique structure of the bile salt micelle is much more tol… more

Solvent Tailoring in Coal Liquefaction. Quarterly Report, February 1982-May 1982. [In Many Cases the Simple Organic Compounds Listed Were Hydrogenated]

Description: Twenty-three model donor solvents were used to rank their efficacy for the dissolution of Western Kentucky No. 9/14 coal. The transfer of hydrogen from the solvent to the coal fragments, as measured by coal conversion, was examined at three levels of available hydrogen. The hydrogen donors are ranked according to their ability to convert coal to THF solubles. Aromatic analogs of the model donors showed little ability to convert coal to THF solubles. Factors which influence hydrogen donation inc… more

Radiolabeled androgens and progestins as imaging agents for tumors of the prostate and breast

Description: We are preparing progestins and androgens, labeled with the single photon emitters technetium-99m and rhenium-186 and the positron-emitting radionuclide fluorine-18. In both cases, ligands selected have very high affinity for the respective receptor, low affinity for blood and non-specific binders and to be reasonably resistant to metabolism: The progestins will be derivatives of the potent progestins ORG2058, norgestrel, RU486, and an unusual retroprogestin and the androgens will be derivative… more

Detection of atypical bile acids in disease states and their identification by gas chromatography-mass spectrometry-computer techniques

Description: The study of the bile acid constituents of serum, bile, urine, and stool of patients exhibiting liver disease has increased in importance with the availability of newer methods for their detection and identification. A cogent question for study has been whether specific bile acids are toxic and thus are the cause of liver disease, or whether they accumulate as a result of disease-induced alteration in metabolism. Examining a wide variety of clinical samples, we have observed that many patients … more

(1) Investigation of the influence of antiarrhythmic agents, such as lidocaine and procainamide, on the chemotaxis and nylon fiber adherence of indium-111-labelled human polymorphonulcear leukocytes (PMNs) in vitro revealed that at normal therapeutic levels of lidocaine and procaine, the adherence and chemotactic function of In-111-PMNs remain unaltered. Results with higher therapeutic blood levels are also discussed. (2) … more
