Immunofluorescence as a Method for the Rapid Identification of Streptococcus Faecalis in Water
The development and refinement of FA has been adequately investigated with major emphasis on pathogenic microorganisms. The development of this technique has reduced both the time and number of biochemical tests necessary to identify a diversity of organisms. The organisms included are the protozoans, as described by Goldman (1953 and 1957) and by Ingram (1961), viruses, as reported by Liu (1955a) and Burgdorfer and Lackman (1960a), pathogenic bacteria which have been investigated by Moody, Goldman, and Thomason (1956), Moody and Winter (1959), Deason, Falcone, and Harris (1957) and Thomason, Cherry, and Moody (1957). Various fungi have been studied with FA by Kaufman and Brandt (1964), Kaufman and Kaplan (1961 and 1963) and Gordon (1958). Therefore, due to the success of the fluorescent antibody technique in many areas of microbiology in previous investigations, the logical assumption was that immunofluorescence might be incorporated into an efficient system in which a specific organism associated with fecal pollution, such as S. faecalis, could be rapidly identified. Based on this assumption, the feasibility of fluorescent antibody techniques, using S. faecalis was investigated as a means of rapid determination of bacterial pollution in water. Although much progress has been achieved in the study of cytochemical reactions by immunofluorescence, no attention has been focused on the application of this method as a determinative tool by which water contamination, due to the presence of the enterococci, could be demonstrated. Specifically, the purpose of the research reported in this dissertation was to devise an applicable, valid, and rapid method that could be employed in the detection and identification of S. faecalis.
Impact of a Genetically Engineered Probiotic Therapy and IGF-1 Genomics in the PAHenu2 Mouse Model of PKU
Absence of functional phenylalanine hydroxylase results in phenylketonuria (PKU). Viable treatments remain few, expensive and secondary conditions such as osteopenia occur in most PKU patients. Objective 1: Given the recently described roles of gut microbes to aid host digestion, an orally administered genetically engineered probiotic as the delivery vehicle for enzyme replacement therapy was created. The engineered probiotic, pHENOMMenal, produced phenylalanine ammonia lyase with significant production of trans-cinnamate (phenylalanine cleavage product) in vitro and resulted in a reduction of 515 μM in blood phenylalanine when fed to PKU animals for 14 days (from 2307µM ± 264µM to 1792µM ± 261µM, n = 6, P < 0.05). The control probiotic produced no change in blood phenylalanine. Thus, pHENOMMenal treatment in PKU mice demonstrated engineered microbes could compensate for a metabolic deficiency of the host. Objective 2: Evaluate the PAHenu2 mouse model of PKU for a genetic discrepancy causing ocular enlargement and delayed development observed only after the PAHenu2 mutation was crossed to the C57BL/6J mouse. When compared to healthy littermates, ELISA indicated a consistent but insignificant decrease in plasma IGF-1 and an increase in ocular IGF-1 in PKU animals. SNP screening demonstrated a differential inheritance of IGF-1 alleles in healthy and PKU animals based on PAH allele inheritance. Ocular and developmental phenotypes in the PAHenu2 colony match those described in previous IGF-1 studies. Understanding the IGF-1 inheritance discrepancy will enable better osteopenia research using PAHenu2 mice and allow breeding of a healthier mouse colony for continued research. Collectively the results from this work describe a new therapeutic approach for treatment of PKU as well as a better understanding of the PAHenu2 mouse model to study this disease.
The Impact of Developmental Stress on Cardiovascular Physiology of Two Archosaur Species: American Alligator (Alligator mississippiensis) and Domestic Chicken (Gallus gallus)
Crocodilians and birds comprise sister taxa of archosaurs, the development of these vertebrates occurs within an egg case that leaves developing embryos susceptible to fluctuations in the nesting environment. Studies suggest that sub-optimal conditions alter morphological growth and cardiovascular physiology. Regulation of the cardiovascular system is immature in the subjects studied, and embryos may rely on humoral rather than neural control of the cardiovascular system. The primary focus of this dissertation was to assess regulatory mechanisms responsible for maintenance of arterial pressure and heart rate. Dehydration stress had marked effects on embryo growth, and altered baseline cardiovascular parameters, while leaving the response to humoral regulator, angiotensin II (Ang II), unaffected. However, dehydrated alligator embryos developed cholinergic tone on heart rate. Hypoxic incubated chicken embryos were reduced in embryo mass, and altered response to humoral regulatory components Ang I and adenosine in addition identifying a novel regulatory component of the cardiovascular response to acute hypoxia. Collectively, these studies add to the existing knowledge of cardiovascular physiology in embryonic archosaurs and suggest that some components of cardiovascular regulation are plastic following developmental stress.
Impacts of the Pyrethroid Insecticide Cyfluthrin on Aquatic Invertibrate Populations in Outdoor Experimental Tanks
The chemical fate and biological impacts of cyfluthrin in aquatic ecosystems were investigated using microcosms (1.9 m^3 concrete tanks) during 1989. Results were compared to a concurrent pesticide registration study using mesocosms (634.7 m^3 earthen ponds). Ten spray drift and five soil runoff simulations were conducted. Pesticide loadings were scaled by system volume, with the same experimental design in ponds and microcosms. Aqueous cyfluthrin concentrations and sediment residue values were generally higher in microcosms, while aqueous half-life was shorter in the smaller systems.

In Vitro Determination of the Cellulose-Decomposing Rates of Twelve Denton County, Texas Soils
In this study twelve types of top soil were collected under aseptic conditions. The cellulose-decomposing rates of these were compared in order to determine the relative rates in the cellulose-decomposing potential of the microorganisms involved. Furthermore, this investigation is designed to acquire pertinent information on the rate at which natural cellulose materials are returned to available plant food.

Induction of 16α Hydroxylase in Human Cultured Lymphocytes
A method is presented for 160hydroxylase (SAH) induction in cultured human lymphocytes. SAH, a microsomal-associated enzyme, effects the oxidative conversion of 17pestradiol to estriol, which competes for cytoplasmic binding sites. 17,-estradiol and estrone are known mammary carcinogens, while estriol and its epimers have been suggested to have anticarcinogenic properties. To substantiate genetic variations of hydroxylase activity, an analysis of estrogen-induced cultured human lymphocytes was conducted to evaluate the frequency distribution of low, intermediate, and high SAH activity. Frequency analysis indicated that the control population distribution of SAH activity does not corroborate a proposed trimodal expansion of human SAH activity. A log normal distribution of SAH activity does exist, which suggests a polygenic mode of genetic control. SAH activity in a population of breast cancer patients and relatives of breast cancer patients showed no statistical difference from the SAH activity in the control population.
Induction of Aryl Hydrocarbon Hydroxylase in Ambystoma tigrinum
Aryl hydrocarbon hydroxylase (AHH) was induced 15-fold in Ambystoma tigrinum by intraperitoneal injection of 3-methylcholanthrene in corn oil, or 10-fold by addition of aromatic polycyclic hydrocarbons to the aqueous environment of the neotene animal. The cytochrome P-450-associated microsomal enzyme is similar to the inducible, one-gene, autosomal-dominant system typical in the laboratory mouse and man. Differences in optimal temperature for enzyme induction and activity were noted in organ culture of human and Ambystoma tissues, and ratios of benzpyrene metabolites differed between Ambystoma and Mus. The half life of enzyme activity induced in vivo was related to the excretion of hydrocarbon metabolites.
Induction of Visible Mutations in Mormoniella by Use of Low Frequency Ultrasonic Energy
Low-frequency ultrasonic energy was utilized in an attempt to induce visible mutations in the parasitoid wasp Mormoniella vitripennis. This study demonstrates that low frequency ultrasound may be used as an effective mutagenic agent in this organism, and suggests that it may have applications to other genetic systems.

Integrating Concepts in Modern Molecular Biology into a High School Biology Curriculum
More so than any other science in the past several decades, Biology has seen an explosion of new information and monumental discoveries that have had a profound impact on much more than the science itself. Much of this has occurred at the molecular level. Many of these modern concepts, ideas, and technologies, as well as their historical context, can be easily understood and appreciated at the high school level. Moreover, it is argued here that the integration of this is critical for making biology relevant as a modern science. A contemporary high school biology curriculum should adequately reflect this newly acquired knowledge and how it has already has already begun to revolutionize medicine, agriculture, and the study of biology itself. This curriculum provides teachers with a detailed framework for integrating molecular biology into a high school biology curriculum. It is not intended to represent the curriculum for an entire academic year, but should be considered a significant component. In addition to examining key concepts and discoveries, it examines modern molecular techniques, their applications, and their relevance to science and beyond. It also provides several recommended labs and helpful protocols.
Integration of field-based analysis of plant community dynamics with quantitative analysis of landscape change in the Ray Roberts Lake area, 1987—1997
This study focused on the effectiveness of integrating traditional plant community analyses with landscape ecological analyses based on remotely sensed data. A temporal analysis of plant community diversity was conducted for major plant communities of the Ray Roberts Lake area using transect monitoring data collected between 1987 and 1997. Landscape analyses were performed with FRAGSTATS*ARC using classified SPOT satellite imagery for 1987 and 1997. Although the methodology developed in this work was exploratory, it was found that characterizing the dynamics of major plant communities in the study area produced a more effective and insightful analysis of Ray Roberts Lake area landscape dynamics.
Investigating the Ability of Pseudomonas aeruginosa pyrE Mutants to Grow and Produce Virulence Factors
Pseudomonas aeruginosa are medically important bacteria that are notorious for causing nosocomial infections. To gain more knowledge into understanding how this organism operates, it was decided to explore the pyrimidine biosynthetic pathway. Pyrimidine synthesis, being one half of the DNA structure, makes it a very important pathway to the organism’s survivability. With previous studies being done on various genes in the pathway, pyrE has not been studied to the fullest extent. To study the function of pyrE, a site directed mutagenesis was done to completely knock out pyrE, which encodes the protein orotate phosphoribosyl transferase that is responsible for converting orotate into orotate monophosphate (OMP). A mutation in this step leads to accumulation and secretion of orotate into the medium. Analyzing virulence factors produced by the mutant and comparing to the wild type, some intriguing features of the mutant were discovered. One of the findings was the over expression of virulence factors pyoverdin and pyocyanin. Pyocyanin over expression, based on the results of this study, is due to the accumulation of orotate while over production of pyoverdin is due to the accumulation of dihydroorotate. The other virulence factors studied were motility assays, exoproducts, and growth analysis. All virulence factor production was reduced significantly in the mutant compared to the wild type. The casein protease assay showed absolutely no production of proteases in the mutant. The conclusion is that orotate accumulation leads to a significant reduction in virulence factor production in Pseudomonas aeruginosa. In addition to that, it was found that excess orotate in the wild type led to a decrease in quorum sensing regulated products.

Although several procedures have been employed for the eradication of tastes and odors from water supplies, none of them are completely successful.
Investigation of Pyrimidine Salvage Pathways to Categorize Indigenous Soil Bacteria of Agricultural and Medical Importance and Analysis of the Pyrimidine Biosynthetic Pathway's Enzyme Properties for Correlating Cell Morphology to Function in All Phases of Growth
This dissertation comprises three parts and is presented in two chapters. Chapter 1 concerns Arthrobacter, a bacterium with an intriguing growth cycle. Whereas most bacteria exist as either a rod or coccus, this bacterium shares the rod/coccus lifestyle. It therefore seemed important to examine the growth regulatory pathways from the rod and coccus. The committed step, that catalyzed by aspartate transcarbamoylase (ATCase), in the pyrimidine biosynthetic pathway was chosen. The ATCase in Arthrobacter is like the well known Pseudomonas enzyme except that it has an active dihydroorotase (DHOase) associated. Included in Chapter 1 is the description of a microorganism, Burkholderia cepacia, whose ATCase has characteristics that are at once reminiscent of bacteria, mammals, and fungi. It differs in size or aggregation based on environmental conditions. In addition, it has an active DHOase associated with the ATCase, like Arthrobacter. B. cepacia is important both medically and for bioremediation. Since B. cepacia is resistant to most antibiotics, its unique ATCase is a prime target for inhibition. Whereas the first chapter deals with the de novo pathway to making pyrimidines, which is found mainly in the lag and log phase, Chapter 2 addresses the salvage pathway, which comes more into play during the stationary phase. This section focuses on the isolation, identification, and grouping of a number of natural soil bacteria from various soil locations. These organisms are important agriculturally, medically, and industrially. Addition of these soil isolates to poor soils has been found to improve the soil. In a previous study by D.A. Beck, the salvage schemes for a number of laboratory strains of microorganisms were determined. Nine separate classes of salvage were designated by determining the salvage enzymes present. Investigations of neuronal network responses to electrical stimulation in murine spinal cultures.
Spontaneous activity in neuronal networks in vitro is common and has been well documented. However, alteration of spontaneous activity in such networks via conditioning electrical stimulation has received much less experimental attention. Two different patterns of electrical stimulation were used to enhance or depress the level of spontaneous activity in spinal cord cultures. High-frequency stimulation (HFS), a method routinely shown to increase the efficacy of synaptic transmission, was employed to augment spontaneous activity. Low-frequency stimulation (LFS), the technique often applied to depress synaptic efficacy, was employed to decrease spontaneous activity. In addition, LFS was used to reverse the effect of HFS on spontaneous activity. Likewise, HFS was applied to counter the effect of LFS. Because these networks were grown on multi-microelectrode plates (MMEPs), this allowed the simultaneous stimulation of any combination of the 64 electrodes in the array. Thus, the possible differences in response to single versus multi-electrode stimulation were also addressed. Finally, test-pulses were delivered before and after the conditioning stimulation on the same stimulation electrode(s) in order to assess the change in mean evoked action potentials (MEAPs). Dissociated spinal tissue from embryonic mice was allowed to mature into self-organized networks that exhibited spontaneous bursting activity after two weeks of incubation. Spontaneous activity was monitored from up to 14 recording channels simultaneously. Although uniform responses to stimulation across all recording electrodes were rarely observed, a large majority of the recording channels had similar responses. Spontaneous activity was increased in 52% of 89 HFS trials, whereas activity was decreased in 35% of 75 LFS trials. The duration of most of these increases was less than 5 minutes. When there were substantial and long-term (> 15 min) changes in spontaneous activity, the opposing stimulation pattern successfully reversed the effect of the previous stimulation. The Isolation and Characterization of a Hitherto Undescribed Gram-Negative Bacterium
A unique undosciribed gramnegative rod is extensively characterized in this study. The cells of this unusual water isolate measure 1.2 X 6.5 microns, The most distinguishing characteristic of the bacterium is a polar tuft of 35-40 flagella that aggregate to function as a single organelle which is visible under phase contrast. Aging cells deposit poly- -hydroxybutyric acid granules which are bound by an inclusion membrane made up of four distinct layers. It also possesses an unusual exterior membrane outside the cell wall which contains large fibrils of protein running at a slight angle to the longitudinal axis of the cell. The guanosine-cytosine ratio was found to be 62.2$. The organism's taxonomic position was further investigated by immunological, morphological, and biochemical methods. It was found to be most closely akin to members of the genus Pseudo onas, although somewhat divergent from other species classified in this genus. After careful evaluation of the findings obtained during this study, the new bacterium was subsequently named Pseudomonas multiflagella.

Isolation and Partial Characterization of Carotenoid Pigments of Mycobacterium Rhodochrous

In addition, it was the purpose to determine whether or not the type of pigments found in M. rhodochrous is typical of those found in other mycobacteria.
Isolation and Partial Characterization of Pigment Mutants of Corynebacterium poinsettie ATCC 9682
Carotenoid pigments were extracted from Corynebacteriuma poinsettiae (wild type) ATCC 9682, and from 108 mutants obtained by exposure of a streptomycin resistant strain of C. poinsettiae to ultra-violet light irradiation and N-methyl- N'-nitro-N-nitrosoguanidine. The pigments were characterized by their absorption maxima, Rf-values, and partition ratios in petroleum ether and methanol. Thin layer chromatography was used to compare pigments of the wild type with those of the mutants. Possible biosynthetic pathways in carotenoid synthesis of the wild type were postulated on the basis of the observed genetic blocks. Mutants were found which suggested the existence of a linear pathway in carotenoid synthesis from the aliphatic C4 0 molecule to the bi-cyclic C50-diol. Other mutants suggested possible alternative pathways in the biosynthesis of these pigments or the presence of intermediates not detectable by thin layer chromatography.
The Isolation, Cultivation and Testing of Organisms Anatagonistic to a Streptomycin Resistant Strain of Pseudomonas Aeruginosa
The problem of finding an efficient antibiotic against Pseudomonas aeruginosa which can be used in the clinical treatment of genito-urinary tract infections resistant to treatment by streptomycin has not yet been solved. Therefore, this problem has consisted of first, the acquisition of possible inhibitors of the streptomycin resistant strain of Pseudomonas aeruginosa; second, the selection and identification of those which show a marked antagonism toward this organism; third, the determination of the antibiotic spectra of the inhibitors; fourth, the determination of whether the streptomycin resistant strain could also acquire a resistance to the antibiotic produced by its inhibitors; and last, an attempt to evaluate the therapeutic possibilities of the antibiotics demonstrated.
Isolation, Identification and Cultivation of Four Phytopathogenic Fungi
This investigation includes a morphological and cultural study of four rather common pathogenic fungi one of which attacks Zinnia elegans, one Lagerstroemia indica, one Ligustrum lodense and one Euonymus japonica. The problem includes, first, a determination of the genera of these four fungi as revealed by their morphology in natural habitat; second, a determination of the growth and cultural characteristics of these on Cornmeal, Potato, Bean, Prune and Carrot agars; third, a comparison of the growth and cultural characteristics of these in natural habitat with the same on the various agars used; and fourth, an evaluation of the five artificial culture media in so far as their usage is concerned in the growth and cultivation of these four species of fungi.

Laboratory and field studies of cadmium effects on Hyalella azteca in effluent dominated systems.
Laboratory single-species toxicity tests are used to assess the effects of contaminants on aquatic biota. Questions remain as to how accurately these controlled toxicity tests predict sitespecific bioavailability and effects of metals. Concurrent 42-day Hyalella azteca exposures were performed with cadmium and final treated municipal effluent in the laboratory and at the University of North Texas Stream Research Facility. Further laboratory testing in reconstituted hard water was also conducted. Endpoints evaluated include survival, growth, reproduction, and Cd body burden. My results demonstrate that laboratory toxicity tests may overestimate toxicity responses to cadmium when compared to effluent dominated stream exposures. Discrepancies between endpoints in the three tests likely resulted from increased food sources and decreased cadmium bioavailability in stream mesocosms
A Laboratory Study of the Asiatic Clam (Corbicula fluminea Müller) as Influenced by Substrate, Food Source and Water Type
Growth of Corbicula fluminea was monitored in the laboratory. Three experiments were conducted. Experiment I utilized three substrates and one food type. Experiment II utilized three substrates and two food types. Experiments I and II were conducted to determine if substrate type or food type effected growth. Experiment III used no substrates, one food type and was conducted to determine growth response to different types of water. Clams were maintained in three substrates: sand, gravel and clay. Clams were also maintained without substrate. Growth was monitored by measuring shell length (mm) and recording the weight (mg) of clams over a period of thirty days. At the end of the test period data were evaluated for normality and homogeneity.
Lactate Dehydrogenase of Hymenolepis Diminuta: Isolation and Characterization
Lactate dehydrogenase was isolated in pure form from crude extract of the cestode Hymenoleois diminuta by heat treatment and column chromatography. The purified enzyme has a specific activity of 106 units per mg protein. The molecular weight of the purified protein was 75,000 as determined by Sephadex gel filtration and analytical ultracentrifugation. An equilibrium ultracentrifugation study suggests a subunit molecular weight of 39,000. From these data, a dimer form of the native enzyme is proposed.
Landscape Ecological Characteristics of Habitat of the Red-cockaded Woodpecker
Geographic information systems (GIS) technology was used to analyze factors influencing habitat use by an endangered species, the red-cockaded woodpecker. The study area was the western part of the Raven Ranger District of the Sam Houston National Forest, Texas. The factors considered included both structural characteristics and spatial relationships among stands of trees in the habitat.

The purpose of this investigation was an attempt to clarify the relatives taxonomic position of and to formulate some of the basic cyclic morphological and physiological processes occurring in an aquatic actinomycete possessing streptomycetal characteristics.
Life History Allocation of Energy to Growth and Reproduction in Gizzard Shad, Dorosoma cepedianum, in North Central Texas
Life history allocation of energy to growth and reproduction was determined for female gizzard shad. Absolute caloric energy allocated to eggs increased with age. The relative amount of production energy directed to reproduction increased with age up to 5 years and then decreased. Seasonal variation in lipids was studied. Quantitative changes occurred in ovarian lipids during the reproductive cycle. Carcass lipids varied seasonally. Age of sexual maturity was 3 years. Delayed maturity is attributed to high allocation of energy to growth enabling shad to outgrow intense competition and predation in pre-reproductive ages; there is little competition and predation in reproductive shad. Growth rates and condition factors indicated constant availability of food seasonally.

The Life History and Ecology of the Mayfly Neochoroterpes mexicanus Allen (Ephemeroptera: Leptophebiidae)
The life history and ecology of Neochoroterpes mexicanus was studied from data collected September, 1971, to August, 1972, and January to December, 1973, in the Brazos River, Texas. Nymphal development, instar analysis, voltinism, standing crops, and production estimates were determined from the quantitative samples taken in 1971 and 1972. Aspects of the life history and food habits of 230 specimens were arrived at from qualitative samples and light box captures in 1973. Laboratory investigation in 1973 helped in establishing instar analysis, egg incubation and description, and first instar descriptions. Neochoroterpes mexicanus appeared to have three generations per year with brood overlap in the summer and fall. It displayed 16 and 19 instars for overwintering and combined summer generations respectively.

Light and Electron Microscope Studies on the Chemotherapeutic Effect of a Combination of Dimethyl Sulfoxide and Hematoxylin on a Transplantable Lymphosarcoma
Investigations concerning the cellular response of tumor tissue to treatment with dimethyl sulfoxide and hematoxylin have not been reported. To establish the response of neoplastic tissue and cells to this combination of agents, this study was undertaken to determine the effects of dimethyl sulfoxide and hematoxylin on a transplantable lymphosarcoma in mice.
Light Spectra Distributions in Temperate Conifer-Forest Canopy Gaps, Oregon and in Tropical Cloud-Forest Canopy, Venezuela
Light spectra distributions were measured in two different montane forests: temperate and tropical. Spectral light measurements were made in different sized canopy gaps in the conifer forest at H. J. Andrews Experimental Forest in Oregon, USA. Researchers at Oregon State University created these gaps of 20 m, 30 m, and 50 m in diameter. In the tropical cloud forest, spectral light measurements were made in two plots that were permanently established at La Mucuy Parque Nacional in Venezuela, in collaboration with researchers at Universidad de Los Andes. In both studies, spectra and distributions of physiologically active light were analyzed: red, far-red, R/FR ratio, and blue light.
Limitation of Primary Productivity in a Southwestern Reservoir Due to Thermal Pollution
Evidence is presented to support the conclusions that (1) North Lake reservoir is less productive, contains lower standing crops of phytoplankton and total organic carbon than other local reservoirs; (2) that neither the phytoplankton nor their instantaneously-determined primary productivity was detrimentally affected by the power plant entrainment and (3) that the effect of the power plant is to cause nutrient limitation of the phytoplankton primary productivity by long-term, subtle, thermally-linked nutrient precipitation activities.
Liver Glyoxalase Activity in Normal Mice and Mice with Lymphosarcoma
It is the purpose of this investigation to determine the variation of glyoxalase activity in liver of normal mice and in the liver of mice bearing a lymphosarcoma and to compare the glyoxalase activity of the lymphosarcoma with values previously reported in the literature for other tumor types. Further, if there is indeed a variation in liver glyoxalase activity between normal and tumor-bearing mice, it will be compared to the variation in the activity of two other enzymes present in liver tissue in relatively high concentration.
