Degradation of Phenolic Acids by Azotobacter Species Isolated from Sorghum Fields

Description: Sorghum plants excrete phenolic acids which reduce subsequent crop yields. These acids accumulate in field soil by combining with soil and clay particles to form stable complexes which remain until degraded by bacterial metabolism. The amount of phenolic acids in soil samples were obtained by gas chromatography measurements, while Azotobacter populations were obtained by plate counts in 40 sorghum field samples from Denton County, Texas. One can conclude that increasing the Azotobacter populati… more

The Effect of Intermediate Filament Inhibitors on Steroidogenesis and Cytoskeleton in Y-1 Mouse Adrenal Tumor Cells

Description: When Y-1 mouse adrenal tumor cells were treated with sodium orthovanadate, an intermediate filament (IF) inhibitor in BHK21-F cells, there was no change in the amount of 20α-dihydroprogesterone produced. A neurofilament inhibitor, β, β'-iminodipropionitrile (IDPN), enhanced the ability of Y-1 cells to produce steroid in response to ACTH by acting on the plasma membrane. Electron microscopy of Y-1 cells extracted with Triton X-100 revealed that both vanadate and IDPN caused the aggregation of cy… more

Effects of Exogenous Steroids on the Adrenal Plasma Membrane Alteration of Steroidogenesis and Cell Morphology

Description: Using cultured Y-1 mouse adrenal tumor cells which produce the steroid 20(-hydroxypregn-4-en-3-one (20-DHP), it was found that 10-5 M corticosterone and deoxycorticosterone increased basal and inhibited ACTH-induced 20-DHP production. The steroid effects were concentration-dependent, reversible, and specific since six other steroids did not stimulate steroidogenesis and varied in their ability to inhibit ACTH-induced steroidogenesis. Cytochalasin D inhibited steroid-stimulated 20-DHP production… more

Ethylbromo Acetate as an Olfactory Probe

Description: Olfactory transduction mechanisms are best studied when specific blocking agents are available with which to modify cellular responses to odorant stimulation. This study is an electrophysiological investigation of functional group-specific olfactory acceptor mechanisms using ethylbromo acetate (EtBrAc), a World War I war gas which inhibits olfactory responses to odorant stimulation. The major findings of this investigation show that (1) vaporous EtBrAc is found to be a quick and effective inhib… more

An Evaluation of the Short-Term Embryo-Larval and Seven-Day Larval Test Methods for Estimating Chronic Toxicity of Zinc to the Fathead Minnow (Pimephales promelas)

Description: Chronic toxicity of zinc to Pimephales promelas was estimated by conducting replicate static and static-renewal short-term embryo-larval tests and static-renewal seven-day larval tests. The two test methods were highly reproducible. Daily renewal of test solutions had little effect on the toxicity of zinc, however, the stage of development at which exposure was initiated affected the sensitivity of the toxic endpoints measured. The most sensitive and reproducible endpoint in the embryo-larval t… more

Factors Inhibiting Dissociation Of Neisseria gonorrhoeae Cells

Description: The initial studies reported in this dissertation were attempts to induce mutations in those genes which control dissociation in cells of Nei sseria gonorrhoeae. These studies led to an investigation of survival curves of cells grown in liquid media. Instead of survival curves reflecting the diploid nature of gonococci, multiple cell kinetics were observed. It was found that large clumps contained a predominance of cells of the T2 type and that when these clumps were dispersed by DNAase, it app… more

Failure of Azotobacter Vinelandii to Fix Nitrogen in Soil

Description: It was found that Azotobacter vinelandii grown in a dialyzed soil medium did not fix nitrogen and had a much lower rate of respiration than when grown in Burk's nitrogen-free medium. When para-hydroxybenzoic acid served as the added oxidizable organic carbon source in dialyzed soil medium, the azotobacter grown in it were found to be unable to fix nitrogen. On the other hand, A. vinelandii fixed nitrogen when grown in soil supplemented with glucose. It was concluded that natural conditions in t… more

Fate of Endothall in Aquatic Environments

Description: Hazard assessment of pesticides in aquatic environments requires accurate predictions of persistence and compartmentalization. A strategy for developing confidence in predictive fate models, such as the Exposure Analysis Modeling System (EXAMS) and the Simplified Lake and Stream Analysis model (SLSA), is to test the models using carefully chosen chemicals in semi-controlled and field situations. An objective of this approach would be to isolate the variability in a particular fate process and t… more
